This is the culmination of >10 years of work by generations of students and postdocs. Many false starts. And we know that others have tried too. Structural insights into the activate-and-release mechanism of NLR activation. And we can engineer it too! Kudos Team 🙏🔥

Check out our latest work! AlphaFold 3 revealed a transient immune receptor complex that eluded biochemical studies for almost a decade🤯 We describe a conserved structural logic underlying sensor–helper communication in an NLR immune receptor network 🧵👇 biorxiv.org/content/10.648…

Can we rewire how plants fight disease? We used AF3 to predict, validate, and engineer a sensor–helper interface that escaped biochemistry for years 🤯 Grateful to @mpcontreras4 & @KamounLab for trusting me to be part of this story. Full story below 👇🏻

Neat use of @GoogleDeepMind AI Co-Scientist and AlphaFold 3 from @amiralito_ @KamounLab @TheSainsburyLab They discovered a massive potato immune protein (an 11-mer resistosome) with Co-Scientist suggesting new structural clues that helped the team hunt this down. Exciting leap forward in decoding plant immunity with implications for global food security.

Protein structure prediction changed biology. Protein assembly discovery is next. We present SNI (Structural Novelty Index), a scalable framework for finding new protein complexes. We used it to uncover an unexpected resistosome assembly: 11-mer. 🧵👇🏻 biorxiv.org/content/10.648…

Just published: Breaking the Pattern with AlphaFold: Introducing the Structural Novelty Index AlphaFold has transformed how we predict protein structures. But sometimes the most interesting discoveries come from asking a different question: which proteins break the pattern? medium.com/p/breaking-the…

UP TO ELEVEN - We expressed potato NRC7, imaged it by negative-stain EM, ran 2D classification. 🔬💥 Rings ~250 Å across — way bigger than canonical hexamers (~150 Å). 11 protomers. (5/n)

We conclude that autoactive NRC helpers function independently of upstream sensors like Prf. In other words, the NRC network is unidirectional—once a helper is (auto)activated, it can induce cell death and does not depend on a sensor NLR.

NEW! A short update on plant immunity - Do autoactive helper NLRs of the NRC phylogenetic clade require the sensor NLR Prf to induce hypersensitive cell death? We think: No The NRC network is unidirectional and autoactive NRC helpers do not depend on sensors for their activities.
