𝐋𝐨𝐧𝐠𝐞𝐯𝐢𝐭𝐲 𝐢𝐬 𝐊𝐢𝐥𝐥𝐢𝐧𝐠 𝐲𝐨𝐮, 𝐣𝐮𝐬𝐭 𝐦𝐨𝐫𝐞 𝐬𝐥𝐨𝐰𝐥𝐲!🧬 Apologies for the rant! A large proportion of the modern longevity community seems to be overdoing it, and simply doing too much and literally killing themselves to stay young, in my opinion! Paralysis by “over -analysis”, has destroyed many a great athlete 🏋️‍♂️and teams over the years, instead of just enjoying the game and playing with a smile on their face, and I feel the longevity community is doing the same, and unfortunately going in the same direction. Take Bryan Johnson, arguably the poster child of the longevity world and credit to him for pushing boundaries (seriously, well done Bryan!). But the level of stress I encounter, both online and in daily conversations, is truly alarming. Questions like: “Should I be tracking this? Should I take that? Do I need cold plunges, saunas, what blood tests, how often? Should I be practicing mindfulness, especially since I’m always stressed?!” The last point sums it up perfectly. So much stress and pressure to look, feel, or live younger may actually be causing more harm than good. Now, you might say, “Hang on James, isn’t your company, @Muhdohealth , in the longevity game with your DNA and epigenetic testing?🧬” And yes, you’re absolutely right. It might sound like I’m a turkey voting for Christmas! But here’s my caveat: Having an understanding on your current health, and a sense of the trajectory you’re on, can be genuinely useful and provide helpful nudges in the right direction. But, and it’s a “big” but, obsessing over every detail of your health can end up accelerating problems rather than solving them. Sometimes exhausting yourself to live longer simply means you’re not really living. 𝐅𝐨𝐫 𝐞𝐱𝐚𝐦𝐩𝐥𝐞, my epigenetic biological age matches Bryan Johnson’s, and I’m also 47 years young! Yet I still enjoy the occasional beer and steak for dinner each night (delicious, by the way). My focus isn’t just adding years to my life but adding life to my years! That’s the real mission behind Muhdo Health, who are flying the flag for British Science🇬🇧 P.S. Sometimes, having a cold beer 🍺does more for your smile and wellbeing than any cold plunge 🥶ever could. 𝐍𝐞𝐰 𝐒𝐭𝐮𝐝𝐲: This might be the most underrated nutrient in the longevity Game! Sorry, it’s not NMN!! 🧬 As much as certain companies and self-proclaimed longevity experts like to promote that NMN is the secret supplement to slow ageing!! To date, the evidence is wishy washy at best! Not to say it doesn’t have some benefit. I’m sure that it does, but as Carl Sagan once said: “𝑬𝒙𝒕𝒓𝒂𝒐𝒓𝒅𝒊𝒏𝒂𝒓𝒚 𝒄𝒍𝒂𝒊𝒎𝒔 𝒓𝒆𝒒𝒖𝒊𝒓𝒆 𝒆𝒙𝒕𝒓𝒂𝒐𝒓𝒅𝒊𝒏𝒂𝒓𝒚 𝒆𝒗𝒊𝒅𝒆𝒏𝒄𝒆” Now, I’m sure you aware of various minerals such as Calcium, Magnesium and potassium. These are known as “Macro Minerals”, as we require slightly larger amounts each day to stay healthy. Then we have another group which is aptly named “Trace Minerals’ as we require these in smaller amounts and includes Iron, Zinc, Copper and Selenium. Now, selenium is an essential trace mineral which isn’t quite as familiar as other minerals such as Magnesium or Zinc, but one which is involved in numerous physiological processes, ranging from antioxidant protection to thyroid hormone metabolism. I’m pleased to say that @Muhdohealth have now had our DNA methylation clock 🧬 validated, published and peer reviewed.📚🔬 👉 lnkd.in/eJFw8FCr 👈 This is a huge stride forward for the team and UK 🇬🇧Science 🔬in general. We were the first DNA and Epigenetics 🧬company globally to launch a biological age and epigenetics app back in 2019 just before the pandemic. And have now validated in many ways our unique science 🔬with the help of Henry Chung from the University of Essex 𝐏𝐫𝐞𝐜𝐢𝐬𝐢𝐨𝐧 𝐦𝐞𝐞𝐭𝐬 𝐬𝐢𝐦𝐩𝐥𝐢𝐜𝐢𝐭𝐲. 𝐀𝐛𝐬𝐭𝐫𝐚𝐜𝐭 𝐁𝐚𝐜𝐤𝐠𝐫𝐨𝐮𝐧𝐝/𝐎𝐛𝐣𝐞𝐜𝐭𝐢𝐯𝐞𝐬: DNA methylation profiles have emerged as robust biomarkers of ageing, leading to the development of “epigenetic clocks” that estimate biological age. Most established clocks (e.g., Horvath’s 353-CpG pan-tissue clock and Hannum’s 71-CpG blood clock) require dozens to hundreds of CpG sites. This study presents a novel saliva-specific epigenetic clock built on 10 sites identified from Illumina MethylationEPIC (850 k) array data. 𝐌𝐞𝐭𝐡𝐨𝐝𝐬: Saliva DNA methylation was analysed from 3408 individuals (age range 15–89 years, 68% male, 32% female, no diagnosed disease) from the Muhdo Health Ltd. dataset (2022–2024), and 10 CpG sites were selected where methylation levels showed the strongest positive correlations with chronological age (Pearson r = 0.48–0.66, p< 1 × 10−20). These CpGs map to genes involved in developmental and metabolic pathways (including ELOVL2, CHGA, OTUD7A, PRLHR, ZYG11A, and GPR158). A linear combination of the 10 methylation sites was used to calculate a “DNA methylation age”. 𝐑𝐞𝐬𝐮𝐥𝐭𝐬: The 10-CpG clock’s predictions were highly correlated with chronological age (r = 0.80, R2 = 0.64), with a mean absolute error of ~5.5 years. Its performance, while slightly less precise than Horvath’s or Hannum’s multi-CpG clocks, is notable given the minimal marker set. It was observed that all 10 clock CpGs undergo age-related hypermethylation. The biological significance of these loci is discussed, along with the potential health and forensic applications of a saliva-based epigenetic age predictor. 𝐂𝐨𝐧𝐜𝐥𝐮𝐬𝐢𝐨𝐧𝐬: This study demonstrates that a saliva-specific epigenetic clock using only 10 CpG sites can capture a substantial portion of age-related DNA methylation changes, providing a cost-effective tool for age estimation.

The “Godfather of Epigenetic Clocks” and the pioneer of the Horvath Clock back in 2013 Professor Steve Horvath from Altos Labs, Gerard Vinyes Parés from Nestlé Health Science and Josep M del Bas from the Universitat Rovira i Virgili. We had a chat about AI, nutrition, health data, genomics, epigenomics, 🧬metabolomics plus a couple more omics that I can’t remember ha-ha.
