A great teaching moment here for genetic design students: NEVER assume any annotation that just says "promoter" and always inspect the elements along with the publication where said genetic part was designed/discovered no matter how long ago the article was written.

Arrg, 3bp messed up my experiments lol. I was having problems getting any expression from this UBQ1 promoter from lotus, and it turns out the promoter is actually promoter+exon1+INTRON where the YAG acceptor is CRITICAL for proper splicing. Wasted 1 month on 3bp; fixed it now. 🤦‍♂️

Doing an initial sweep of the blue substance that oozed out of the #BlueSoup swab sample. I rung out the swab in DH2O and spun down twice to remove debris. Interesting peak at 590nm! I was using polystyrene cuvettes so disregard the left-most side of the curve. Will quartz later!

Slightly frustrating aspect of agro rhizogens K599 strains is the clumping in rich media (LB). Need to find a nice minimal media like that used for tumefaciens LBA4404. Slower, but less aggregation which hampers plant transformation efficiency. Every strain's a different beast!

Surprisingly good brushless shaker for the price. You have to use these rubber bands that look suspiciously like hair ties, but you can easily gecko-tape a sheet of steel for mag mounts. Fairly quiet too. 40-200RPM. Capacity 2Kg. $130. a.co/d/3IcAj8u
