Advanced Prostate Cancer Consensus Conference
Obesogenic High-Fat Diet and MYC Cooperate to Promote Lactate Accumulation and Tumor Microenvironment Remodeling in Prostate Cancer out on @CR_AACR aacrjournals.org/cancerres/arti… Study explores how systemic metabolism, particularly through a high-fat diet (HFD) rich in saturated fats, accelerates c-MYC-driven #ProstateCancer progression by rewiring tumor metabolism. In a mouse model🐭the combination of c-MYC🧬 activation and an obesogenic HFD led to increased glycolysis and lactate accumulation in tumors, which promoted immune changes, including higher infiltration of tumor-associated macrophages and regulatory T cells. These alterations contributed to tumor growth, therapy resistance, and angiogenesis, which were reduced by lactate dehydrogenase inhibition. Deconvolution models uncovered ⬆️ proportion of M2-like macrophages in ⬆️ glycolytic prostate cancer from patients with ⬆️ saturated fat intake. This strongly suggests that acquisition of glycolytic features contributes to shaping a TME supportive of cancer progression. (11/17)
