Single weekly dose = higher Cmax (peak concentration)
- Triggers appetite suppression harder
- Higher risk of peak-driven side effects (skin sensitivity, GI, HR increases)
- End-of-week troughs may cause appetite rebound
Micro/split-dosing = lower Cmax, similar steady-state AUC
- Smoother appetite control across the week
- Better tolerability - fewer peak-driven side effects
- Glycemic control preserved (driven by AUC, not Cmax)
The trade off is blunted appetite punch for most.
Cmax drives appetite suppression. AUC drives glucose control.
Pick your dosing strategy based on which lever you need to pull harder.

Has anyone found pinning your full weekly amount of Retatrutide in a single dose to be more beneficial than breaking the dose up into two? I’ve seen a lot of discussion around this. Someone was even saying that breaking the dose into two can make your body get used to the dose quicker causing it to be less efficient, thus having to up the dose. Reta summer is officially upon us!
