Engineered enzymes open up so many creative lab techniques. A group out of Sweden created an intentionally error-prone "sloppymerase" that introduces mismatches downstream of single-stranded breaks. SSBs can be located & quantified with high throughput! doi.org/10.1038/s41467…

Molecular evolution is just endlessly fascinating to me. A cool new @CellCellPress paper shows that the eukaryotic mismatch repair machinery has to compete with DNA-binding transcription factor proteins, increasing the mutation rate at TF binding sites. cell.com/cell/abstract/…

Here's a really cool new preprint out of @TheCrick. @LastEgoret al. used DNA fluctuations in an optical trap to estimate DNA intramolecular friction. Bending friction dominates at sub-kbp length scales, deviating from an ideal elastic rod. biorxiv.org/content/10.110…

Interesting paper out of Analytical Chemistry! Click chemistry (azide + DBCO) can be used to greatly accelerate toehold-mediated strand displacement in the presence of short toeholds or mismatches—essentially making the toehold-binding step irreversible. doi.org/10.1021/acs.an…
