In summary, TNF-TNFR2 signal sustains Tregs not through a pure autocrine mechanism. Rather, two Treg subpopulations exist that preferentially express TNFR2 or TNF, which cooperate in enforcing Treg suppression.

🔬🔍Exploring Immunometabolism with Giuseppe Matarese, Claudia Mauri and David Finlay at #ECI2024! 👉Here’s what you need to know: ➡Today's metabolic overwork disrupts the natural oscillations in Treg proliferation, which is crucial for the development of autoimmune diseases and allergies. 🟢The metabolic needs of Breg cells involve a heavy reliance on OXPHOS and the crucial role of thioredoxin in controlling mitochondrial ROS. SLE patients lack Bregs, but thioredoxin treatment can restore this regulatory compartment. 📌Studying immune cell metabolism in vivo presents challenges. A new approach using click chemistry allows for multiplexed metabolic flux and nutrient uptake analysis at single-cell resolution.
