@ATinyGreenCell Original eha105 had pPH1JI in it conferring gent and spec resistance. Derivatives cured the plasmid but in general yeah spec is annoying.

Spectinomycin is the bane of my existence. My EHA105 agro cells are now spontaneously resistant to spec (checked with controls and ruled out dead spec stock). Was trying to transform with specR plasmids and got lawns. Zilch when on kan. Rif resistant. PCR pos for Agro. BAHHHH. 😡
