ALBAN #ESMO25 (atezolizumab/BCG vs BCG in MIBC) @Annals_Oncology did not ⬆️ EFS (HR0.97) unlike sasalimab & durvalumab in a similar setting. The drugs seem similar in metastatic UC so was it trial design making it different from other ICIs? EFS was defined differently.

@tompowles1 The dose intensity for cisplatin in the dd-mvac regimen is at least 25% higher than in the ddp-gem arm. No true comparison is therefore possible

It’s difficult to know what to do with VESPER results for neoadjuvant DDMVAC x6 in UC. While the data encouraging it’s statistically negative. Its therefore hard to see it as practice changing from a purist perspective. It may have been +ve with different designs etc but..#ASCO23

FORT-1 tested rogaratinib vs chemotherapy in FGFR1/3 +ve(RNA) platinum refractory UC. 600 patients screened 175 randomised (60% biomarker +ve?) Efficacy results showed regimens were very similar. Erdafitinib has a better biomarker plan. Randomised trials with erda may still be OK

Summary of 1st line advanced UC IO/chemo trials. IO can’t beat chemo and the PD-L1 biomarker doesn’t help. Chemo/IO (361,130) and Ipi/nivo in PD-L1+ve (901) were disappointing. 3 chemo/IO studies awaited for final OS, but existing data isn’t good. Still a chance for tremilimumab?

Everolimus did not show a significant DFS or OS advantage in adjuvant RCC. This study probably draws a line under the investigation of mTOR (TORC1/2) in RCC and draws a line under adjuvant targeted therapies in RCC. Everolimus role is relatively limited. #ASCO22
