The first paper of this year, “Engineering self-deliverable ribonucleoproteins for genome editing in the brain”, is officially out @NatureComms. This is the first project of my postdoc @doudna_lab, where we focused on delivery method development for CRISPR genome editors in the ribonucleoprotein (RNP) format, with our final goal on in vivo genome editing in the brain. nature.com/articles/s4146… To endow CRISPR genome editors with self-delivery capability, we incorporated cell-penetrating peptides (CPPs) into Cas9/Cas12 proteins to trigger effective cellular uptake and desired genome editing. Upon systematic screening and engineering of Cas9-CPP fusion constructs, we established a C-terminal Cas9 fusion with a peptide derived from human semaphorin-3a, which could promote RNP delivery to neuronal cells in vitro and in vivo with high efficiency. When injected directly into the mouse striatum, these self-deliverable RNPs could generate robust edits in clinically relevant genes. We believe that self-deliverable RNPs would provide a facile and effective platform for genome editing applications in vitro and in vivo, especially for treating genetic disorders in the central nervous system. The news from people’s park this morning truly makes me so sad. I remember going to community events here since I was a kid. The park is one of the only open green spaces in the south side of berkeley and was literally designated as a historic landmark by the city in 1984. (1/)
