Happy to share the results of @RehamAlsaadi_ & @Mizala who uncovered a really cool mechanism linking Salmonella infection to the suppression of #autophagy receptor FAM134B.

The ER-phagy receptor FAM134B is targeted by Salmonella Typhimurium to promote infection dlvr.it/T4q5SZ

We are excited to share this comparative dataset of selective and bulk #autophagy regulators. Thanks to @maximewc for the collaboration, which created a new research focus in the lab. Comments or questions welcome.

@Russell_BioLab Ill try to normalize growth and whatnot as this was a quick and dirty prep of cells taken aroun 20hr make from overnight. Seems very strain dependent since turbos were def in late stationary phase at that point and still wildly competent taking in 1uL of 100ng/uL miniprep plasmid

Heyyyy, my super quick competent cell test using all my ecoli strains gave some fun results. All except BL21 worked. 1. Grow culture overnight 2. Spin down 1mL 3. Resuspend in 100uL TSS buffer 4. Use 10uL of cells per heatshock Even stubborn NEB 5a worked! K-12 amazingly comp!

@maximewc @JohnDoench @GEMb_uOttawa Agreed, arrayed glycerol would be the best for long term usage and targeted application(primarily using a subset of cassettes). Counterpoint, for an individual lab that wants to dabble, arrayed plasmid DNA (~10rxn) would lower the barrier to start.

@JohnDoench @GEMb_uOttawa @Russell_BioLab I think arrayed glycerol stocks would be the most straightforward? Most of our projects are custom KO/KI or transgenic constructs/cell lines, so having a toolkit at the ready for various "asks" would be great
