Today’s KRAS win didn’t start today. It took decades. Daraxonrasib nearly doubled survival in pancreatic cancer. That’s extraordinary! But lets zoom out for a sec: ▪️KRAS was called “undruggable.” ▫️NIH quietly poured ~$172M behind it. ▫️Funding grew 9x. 💡One insight (Shokat lab, 2013) changed everything. This is how breakthroughs are supposed to work: public money → basic insight academic labs → first leap industry → drug + survival curve Today’s headline has MANY authors. @LocasaleLab My wife was on this trial. Stage 4 panc cancer, 40 years old. She was in this trial and lived 18 months after chemo stopped working. We took trips to Spain, Disney, Jersey Shore. She got two more birthdays with our kids. Shove your hype and misrepresentation up your ass.

The $PFE/ $VALN Lyme vaccine was a product for which there was a lot of hope. Results today show OK efficacy. Vaccine efficacy landed at 70%; investors hoped for 80% but thought as low as 60% would be relevant. But the bigger issue is a statistical one. The main goal of the study included a specification on the lower confidence interval -- basically an estimate of the lowest the efficacy could possibly be -- and it missed. That could lead FDA and other regulators to demand another trial. The companies say they will submit anyway. But that's why Valneva shares are down 30%+. Given all the dynamics around RFKJr, the FDA, and the CDC, this will be a very interesting story to watch. I just updated our story with a quote from the noted vaccine expert Kathy Edwards. She calls the study "encouraging news" but said the study was "underpowered and the confidence intervals are wide." She's not sure what regulators will do. statnews.com/202623/lyme-di… via @statnews
