It’s hoped that the resulting agent, [18F]NP3-627,  could enable the clinical development of drugs acting on the NLRP3 inflammasome, lead to a better understanding of their target engagement and optimal dose, and help stratify patients and monitor disease progression.

Using pyridazine-based inhibitors as a starting point, the research team leveraged their improved permeability and brain-penetrant properties to develop imaging agents for NLRP3 that could help measure both target expression and occupancy by a drug in the CNS.

NP3-253 is expected to be a useful tool to further study NLRP3 inhibition, especially in the central nervous system (CNS) in neuroinflammatory conditions.

NP3-253 displayed good efficacy and tissue & brain penetrance. The study also elucidated the molecule’s mechanism as a  NLRP3 inhibitor.
