@joachimgoedhart I used a script that rotates the image from -30 to 30 and checks each time for the sum in horizontal and vertical direction. Idea: when points align, there should be a peak. I you want I can message you the script.

It is easy to manually rotate the dots to align them with the horizontal and vertical axis of the image. Out of curiosity, is there an automatic way to do this (preferably in Python or ImageJ)?

While the term “throughput” is abundantly used, its definition in biomedical settings is not clear. I had the pleasure to discuss the issue with leading scientists in flow cytometry, high-throughput screening and sequencing. More info @naturemethods: nature.com/articles/s4159…
