99% pure what, though? Purity tells you the vial is clean. It doesn’t tell you it’s the right peptide. That’s a completely separate test — and it’s the one that catches the failures purity can’t see. 🧬 🧪 What identity testing does Every peptide has a molecular weight set by its exact amino acid sequence. Change the sequence, add or drop a residue, and the mass changes with it. That mass is a fingerprint. Mass spectrometry measures it. On a COA you’ll see it listed as MALDI-MS. The instrument measures the mass, you compare it to the mass the sequence should produce, and: — match → it’s the peptide on the label ✅ — mismatch → you’ve got something else entirely That’s why a real COA lists “Identity” on its own line, separate from purity, with its own method. ⚠️ Why this matters more than it sounds Picture two vials. Both 99% pure by HPLC. Both clean, single-peak chromatograms. On a purity report alone, identical. Run mass spec: one hits the expected mass, one doesn’t. Same purity. Different molecule. Without the identity test, you’d have shipped both as if they were the same thing. A clean trace of the wrong compound looks exactly as good as a clean trace of the right one. Purity can’t tell them apart. Identity can. 🔍 The honest caveat Mass confirms mass — not the exact sequence order. Two peptides with the same building blocks in a different order can share a mass (called isobaric species). For real-world QC, a correct mass + a clean purity trace is strong confirmation. Full sequence-level proof takes an extra step like tandem MS. A vendor who knows that difference is one worth trusting. "99% pure." Pure by what method? That question ends most conversations in this industry. The number is on the product page. The measurement behind it usually isn't. Four separate questions get asked of a research peptide, and passing one tells you nothing about the others. 🧪 IDENTITY — is it the right peptide? Mass spectrometry measures molecular weight against the expected value. Without it, a vial can be immaculately pure and still be the wrong compound. ⚗️ PURITY — is it one thing, or a mixture? Reverse-phase HPLC separates what's in the vial and measures the peaks. A clean chromatogram means a single dominant compound instead of a soup of truncated fragments left over from synthesis. Note what this number is a percentage of — the peptide fraction. Not the vial. ⚖️ CONTENT — how much peptide is actually in there? This is the one nobody asks about. Purity is a ratio; content is a quantity. Powder in a vial is peptide plus water plus salt. A 99% pure vial can still hold meaningfully less peptide than the label says, and only HPLC quantitation tells you which. 🔬 ENDOTOXIN — is it clean? Bacterial contamination doesn't show up on any of the tests above. A vial can be pure, correctly identified, correctly quantified, and still carry endotoxin introduced during manufacturing. Separate question, separate test — USP <85>, reported in EU/mL. Over the next few weeks I'm breaking each of these down — what the test measures, what the number means, what it doesn't, and how to read a COA without taking anyone's word for it. Including mine. Straight answer on what we run: identity by MALDI-MS, purity by RP-HPLC at 214 nm, content by HPLC quantitation, endotoxin by USP <85>. Every line with the method named. We don't test heavy metals or residual solvents, and I'm not going to write posts implying we do. Part 1 this week: what a purity percentage actually measures. All products research use only. Preliminary safety concerns should always be taken seriously, but so should the evidence before drawing conclusions. I’d like to see the complete chain of documentation: • Photos of the actual vial tested • Vendor label and batch number • Matching batch number on the COA • A COA from an established laboratory • A result that can be independently verified through the lab • Confirmation using an appropriate confirmatory analytical method, not just an initial screening result Until then, I think it’s reasonable to remain cautious without assuming guilt. Transparency protects both researchers and reputable vendors.

Another Fentanyl Positive-or is the Lab Only Now Looking? A second product has returned a preliminary result through ILS, raising questions about when routine fentanyl screening began and how reliable the initial test actually is. We now have a second, separate incident involving a product testing preliminarily positive for fentanyl through ILS, this time Glutathione 1500. I have no further information at the moment beyond this announcement, but the first question that comes to mind is: how long has ILS had a policy of routinely testing every sample for fentanyl, and could that help explain the sudden appearance of these results? If this screening was only introduced recently, we may not be seeing a sudden rise in fentanyl contamination. We may simply be seeing the consequences of the lab only recently starting to look for it. It is all fair and good offering additional testing for free, but if the initial screening method has a high false-positive rate, releasing preliminary results before confirmatory testing serves no one. Vendors are immediately placed under suspicion, customers are frightened and rumours spread long before the official COAs are available. Precaution is important, but so is context. ILS needs to explain when routine fentanyl screening began, how many samples have been screened, how many preliminary positives have occurred and what confirmatory method is used before any result is treated as genuine.

An address that also lists BALI Classic Massages, equipment images lifted straight from manufacturers’ catalogues and more suited to a peptide synthesis and purification workflow, and a domain registered only a few months ago. During yesterday’s FDA Pharmacy Compounding Advisory Committee meeting, PCAC member Dr Melissa Loseke said: “Just last week, I had a patient whose research-grade whatever that they got from wherever was laced with MDMA and ecstasy. That’s not protecting the American people.” I wonder which laboratory tested this mysterious peptide, whatever it was, whether there is a report we can see but most importantly, how on earth the MDMA got there? 👀👀

UPDATE: 6 FOR 7. Ahead of the meeting, FDA staff briefing documents recommended against all seven. The advisory committee didn’t follow suit. Over July 23–24, the Pharmacy Compounding Advisory Committee voted to recommend 6 of the 7 peptides for the 503A Bulks List: ✅ BPC-157 (8–6–1) ✅ TB-500 (8–6–1) ✅ KPV (8–6–1) ✅ MOTS-c (7–5–2) ✅ Semax (recommended) (Couldn’t find the vote count for this one) ✅ Epitalon (7–5–1) ❌ DSIP / Emideltide (6–7–1) The vote is advisory and non-binding. FDA issues the final determination, and any change to the list still goes through a formal rulemaking process. Nothing about these compounds’ current legal status changed today. One honesty note so you don’t post a wrong number: MOTS-c passed on a 7-to-5 vote with two abstentions (7–5–2), and that was the narrowest of Thursday’s four. For Semax, the reporting I can find confirms it was recommended but hasn’t published the exact tally yet — the official roll call apparently isn’t fully up. So I left it as “recommended” rather than invent a split. If you want a fully consistent look, you could drop the parenthetical counts entirely and just use ✅/❌, or wait for the official record and add Semax’s number then. Everything else is confirmed: BPC-157 and KPV passed 8-6 with one abstention; on Friday the committee backed epitalon and semax and rejected emideltide; emideltide failed 6-7 with one abstention while epitalon passed 7-5 with one abstention.

FDA's own scientists went 0 for 7. Ahead of this week's Pharmacy Compounding Advisory Committee meeting, FDA posted briefing documents for BPC-157, TB-500, KPV, MOTS-c, DSIP, Semax and Epitalon. Every single one proposes the same conclusion: do not add to the 503A Bulks List. The committee votes July 23–24. It can disagree — and its recommendation is non-binding either way. "99% pure." Everyone quotes the number. Almost nobody explains what it measures. Here's what that percentage on a peptide COA actually tells you — and the part it quietly leaves out. 🔬 📈 Where the number comes from Purity is measured by HPLC. A tiny sample gets pushed through a column, and different molecules come out at different times. A UV detector records each one as a peak on a graph — the chromatogram. The target peptide is the big peak. The small peaks around it are impurities left over from synthesis: deletion sequences, truncated chains, incomplete products. The math is just: main peak area ÷ total area of all peaks × 100 So "99%" means the target peptide is 99% of the peptide material the detector saw. It's a ratio — the main peak against everything else. 🌈 Why 214 nm matters Look at a real COA and the method reads RP-HPLC (214 nm). That wavelength isn't random — the peptide bond itself absorbs UV right there, so the detector sees essentially any peptide regardless of sequence. But that also defines its blind spot. ⚠️ The part nobody mentions HPLC at 214 nm only sees things that absorb at 214 nm. That's peptides. It does NOT see water, salts, TFA counter-ions, or residual solvents — none of that absorbs there. It's invisible to the detector. So a vial can be 99% pure and still carry non-peptide weight. Purity tells you how clean the peptide is. It says nothing about how much peptide is actually in the vial. Two different questions, two different tests — and content gets its own post later in this series. ✅ How to actually use this Next time you see a purity number, ask: — Is there an actual chromatogram, or just a number typed on a page? — Is the method named? "99%" means little. "99% by RP-HPLC at 214 nm" is a measurement. — Is it batch-specific and run by an independent lab? It doesn’t mean the darkness is gone but there’s an awareness and I’ve got more tools to deal with life’s challenges. I wouldn’t trade sobriety for anything. It’s a better life overall.

One question I haven’t seen answered yet: What exactly was the sample supposed to be, and who submitted it for testing? Context and chain of custody matter when interpreting any unexpected result. A confirmed positive for fentanyl is obviously significant, but understanding where the sample originated is just as important.

Janoshik Provides More Information About the Positive Fentanyl Result Following the positive fentanyl report for the liquid believed to be 5-Amino-1MQ, Janoshik was asked for more information about the testing. He explained that his laboratory used two different immunoassays: one was a pharmacy test strip, while the other was a test used by local law enforcement. According to Janoshik, the law-enforcement test is legally binding until proven otherwise. The sample was not tested just once. Both assays were run twice. The liquid was also diluted in phosphate-buffered saline, yet all four tests returned positive results. Janoshik said his laboratory had previously encountered situations where one fentanyl assay returned positive while another returned negative. However, that was not what happened here: both types of test strip returned positive on both runs. When asked whether 5-Amino-1MQ had previously been tested for fentanyl, Janoshik said not that he was aware of, although he could not search his records conclusively.

What this actually decides — and what it doesn't: The 503A Bulks List is what lets a compounding pharmacy prepare a substance against an individual prescription. That's the question on the table. What's NOT on the table: a ban. All seven already came off FDA's Category 2 restricted list in April. That's what teed up this review in the first place. So the vote is about whether a legal channel opens — not whether one closes. And a yes vote isn't the finish line. Adding a substance to the list requires formal notice-and-comment rulemaking, which doesn't happen in weeks. Real stakes. Slow clock. Nothing changes this week.

I had the same question about a reconstituted vial being tested. I do send in blind studies at times but standard testing I always have a photo of my product with my label and batch number. Where did you find this? I know some labs that do peptide testing have now added Fentanyl testing as an option, makes me wonder why and what caused them to add this. Typically people don't just come up with a random safety idea, sadly it's usually based in something negative happen to trigger it.
