@drnaomi I've seen a hoarder house first hand (and the fallout after the fire department needed to respond). The one I saw was nothing but old newspapers, magazines and actual trash. Hoarding has serious mental illness implications, rich or poor.

Polycyclic tetramate macrolactam genes are common in Streptomyces. But don't confuse that with boring. Check out our recent work if you are interested in orphan genes, BGC mosaicism, and lingering challenges in translating genes to molecules journals.asm.org/doi/full/10.11…

@SubterraFleur Seems about right for a vacation with two kids.

@wildtypeMC You might have to reach out to the chair of Stanford Biochem to track the disposition of Dale's strain collection.

@ATinyGreenCell Actinomycete PCR is challenged by high G+C DNA. Our colony screens are done using cells ground in DMSO (adapted: doi: 10.1016/s0167-7012(02)00235-x.. 2003 Jun;53(3):401-3.J Microbiol Methods). Then PCR is done using FailSafe Premixed buffers.

It worked! Kinda too well and got some secondary amps. No issue I'll just cut that out during digest. Purifying now!

Alright, giving this citT citrate antiporter gene another Q5 PCR attempt. This time testing with and without DMSO (10% v/v final). If this doesn't work then I'll just do nested PCR. I'm getting this gene out come hell or high water, lol.

@wwwplumcom Like others have said, this is most likely an actinomycete. To obtain even growth, use a sterile, low volume mortar and pestle and grind to an even suspension- then save directly as a cryostock or use to sub culture. We do this all the time with environmental isolates.

#microbiology Twitter, any ideas what to google for this phenomenon? when bacteria form hard-to-break-apart balls in the bottom of liquid cultures? I think I'm going nuts.

@BiologyCarly Undergrad story...Doing dozens of West Nile Virus Crow necropsies when my fellow tech cuts straight through a hemorrhaged skull and into her finger with bone shears. With no cure, it was "go wash up and let us know how you feel...". It got real quiet from there.

@academic_exit I went from post doc to industry then back to academia. While I think I know what you are saying, I’m unsure it’s quite that simple for everyone everywhere.
