@drnaomi I've seen a hoarder house first hand (and the fallout after the fire department needed to respond). The one I saw was nothing but old newspapers, magazines and actual trash. Hoarding has serious mental illness implications, rich or poor.

@ATinyGreenCell Actinomycete PCR is challenged by high G+C DNA. Our colony screens are done using cells ground in DMSO (adapted: doi: 10.1016/s0167-7012(02)00235-x.. 2003 Jun;53(3):401-3.J Microbiol Methods). Then PCR is done using FailSafe Premixed buffers.

DMSO is my lord and savior. Ladder Genomic Amp Genomic Amp Genomic Amp + DMSO Genomic Amp + DMSO Genomic Control It worked! Kinda too well and got some secondary amps. No issue I'll just cut that out during digest. Purifying now!

@wwwplumcom Like others have said, this is most likely an actinomycete. To obtain even growth, use a sterile, low volume mortar and pestle and grind to an even suspension- then save directly as a cryostock or use to sub culture. We do this all the time with environmental isolates.

#microbiology Twitter, any ideas what to google for this phenomenon? when bacteria form hard-to-break-apart balls in the bottom of liquid cultures? I think I'm going nuts.

@BiologyCarly Undergrad story...Doing dozens of West Nile Virus Crow necropsies when my fellow tech cuts straight through a hemorrhaged skull and into her finger with bone shears. With no cure, it was "go wash up and let us know how you feel...". It got real quiet from there.

What is your most unhinged story from grad school? Mine was probably the time I picked a dead flounder up off the lab floor and then ran to a public bathroom with it in my hands and sobbed over its body in a stall.

@academic_exit I went from post doc to industry then back to academia. While I think I know what you are saying, I’m unsure it’s quite that simple for everyone everywhere.

When I moved from my post-doc to my first industry role, my base salary went up by more than 300%. Do you love your academic topic enough to under-earn that much? Or want to earn enough that you don't have to think about money every week at the grocery store? Your choice.
