@ChenxinLi2 Great images! Were the flashlights pretty easily obtainable? I found ones that work well with my favorite GFP, would be great for other FPs as well.

@BenMansfeld I agree re: not going cheap! New Gilson's are easy on thumbs like Rainin LTS. Eppendorf's are nice, too. Problem with Rainin is being trapped in their tip ecosystem. Definitely demo, and think about adjustable spacing multichannels to maximize throughput!

@aplantbiologist That's what my team has experienced. Worked with GV3101 lots before LBA4404 and was surprised by the latter's clumping behavior.

Does LBA4404 Agro typically clump & aggregate in culture? I’ve been growing it in YEP or LB media and I can’t seem to avoid the bacteria from clumping together which makes taking an accurate spec reading difficult. Any thoughts?

@ChenxinLi2 I had a rude awakening when I realized my TuMV-GFP and eGFP constructs didn't work equally well under 390 nm UV lamp. Turns out viral GFP label excites at 390 nm while eGFP excites at 488 nm. I now check all my FP sequences on fpbase.org

@DLinYZ I see. I will have to look into that. I have the standard eGFP, I will need to look into what filter and wavelength the scopes and flashlight is.

Tweeps, I can see GFP signal pretty well on a slide with 10x to 20x objective lens, but not strong enough to see it using a flashlight or dissecting scope. Any tricks?

@ChenxinLi2 There are a lot of GFP variants with slightly different excitation/emission spectra. Are the excitation wavelength settings the same for 10-20x scope, dissecting scope, and flashlight?

The experiment was transient expression with agrobacterium infiltration.

@ChenxinLi2 Great question. Looks like in lieu of including a clamp, polymerases in PCR can be fused to sequence non-specific DNA binding proteins: bit.ly/3pp2o4P + clamp seems to be orders magnitude better processivity, tho

Here's a question I wonder about: does DNA polymerase in PCR has processivity? Without DNA clamp, instead of continuous synthesis along the template, does the polymerase put down a few nucleotide & dissociate, then another polymerase molecule comes in and put down another few?

@byu_sam Years ago at Berkeley I was bamboozled into majoring in "Genetics and Plant Biology", not realizing what I actually wanted at the time was MCB w/ emphasis on genetics. Happily still studying plant biology many years later, though!

Doing a survey of how other universities have structured undergrad majors. Very surprised that I am 0/4 in finding a 'genetics' undergraduate major. Many have a mol/cell biology major. But so far no 'genetics'.
