🚨 New paper out in Advanced Science!
We used shear-activated nanoparticles to deliver nitroglycerin to brain collaterals — improving stroke outcomes without systemic side effects.
🧠💥 Targeted therapy for ischaemic stroke is here.

Really enjoyed participating in this great event!! Thanks to @NSWCVRN rising stars sub-committee for organising. Stoked to have won the EMCR category. The quality of the pitches from all participants was outstanding!

3. These findings strongly suggest that rapamycin may be a useful therapy for improving #microvascular #reperfusion and be a an adjunct treatment to improve the effectiveness of recanalization therapies in ischaemic stroke.

2. In mice undergoing #MCAO and reperfusion, rapamcyin treatment significantly increased the diameter of #capillaries at pericyte locations as well as increased the proportion of capillaries that were patent (open) vs those completely constricted (closed) at pericyte locations.

Excited to share that our work showing the #mTOR inhibitor #rapamycin (Sirolimus) can reduce #brain #pericyte constriction following #stroke, is now published in Translational Stroke Research! link.springer.com/article/10.100…

Numerous pharmacological studies modulating pericyte contraction are currently being published! 👀
This interesting @biorxivpreprint shows that #Rapamycin (#mTOR inhibition) relaxes #pericytes, mitigating capillary dysfunction after stroke reperfusion 🧠

@Ella_Maru we assessed the effects 30min after reperfusion, so not sure what the long term effects would be. However colleagues have shown that activation of mTOR out past 4 weeks post-stroke is beneficial. So timing of rapamycin adminsitration is critical.
English

@DocBeardface87 Exciting development! Are there any observed long-term structural changes in the microvasculature post-treatment with rapamycin?

Excited to share our new preprint showing that the #mTOR inhibitor #rapamycin (Sirolimus) can reduce #brain #pericyte constriction following ischemia in vitro and #MCAO #stroke in vivo. biorxiv.org/content/10.110….

@MattDun17 We haven’t, but should. We focused on sirolimus as it has the most evidence behind it in terms of a cerebrovascular protective effect in stroke models. However, there is emerging evidence of the benefits of everolimus in stroke models also.
