3. These findings strongly suggest that rapamycin may be a useful therapy for improving #microvascular #reperfusion and be a an adjunct treatment to improve the effectiveness of recanalization therapies in ischaemic stroke.

2. In mice undergoing #MCAO and reperfusion, rapamcyin treatment significantly increased the diameter of #capillaries at pericyte locations as well as increased the proportion of capillaries that were patent (open) vs those completely constricted (closed) at pericyte locations.

@Ella_Maru we assessed the effects 30min after reperfusion, so not sure what the long term effects would be. However colleagues have shown that activation of mTOR out past 4 weeks post-stroke is beneficial. So timing of rapamycin adminsitration is critical.

@MattDun17 We haven’t, but should. We focused on sirolimus as it has the most evidence behind it in terms of a cerebrovascular protective effect in stroke models. However, there is emerging evidence of the benefits of everolimus in stroke models also.

4. These findings strongly suggest an anti-contractile effect of rapamycin in vivo following stroke and thus rapamycin may be a useful therapy for improving microvascular reperfusion following recanalization therapies in ischaemic stroke.
