We identified a new member of the NEDD4 family in Drosophila, Hecw, and we generated catalytic inactive and KO mutants to find that lack of Hecw ubiquitination activity results in defective oogenesis, premature aging and climbing defects associated with neuronal loss. 2/4

Finally, Fmrp-regulated mRNA targets are aberrantly expressed in oocyte (Orb, an oocyte determinant) and neurons (Profilin, a causative familiar ALS gene). This last event is particularly intriguing as it opens new possibility for the treatment of this devastating disease. 4/4

The biophysical properties of germ and neuronal RNPs are affected, they become larger and transition from a liquid to a less dynamic gel-like state. We dissected the mechanism and identified the Hecw-relevant target, Fmrp, a conserved RNA-binding translational repressor. 3/4
