9/ Overall, our results show that simultaneous disruption of citrate export and NA compartmentation leads to major alterations of the ionome, impaired metal partitioning, persistent oxidative stress and defects in RAM function.

8/ While all the other genotypes produce siliques under hydroponic conditions, the double mutant fails to complete reproductive development. Seeds were only obtained from soil-grown plants supplemented weekly with sequestrene, and they show elevated Fe and Zn levels.

7/ These phenotypes are associated with strongly reduced root-to-shoot translocation, despite the pronounced induction of the NA biosynthetic pathway, which likely reflects an attempt to restore metal mobility.

5/ We observed strong accumulation of Fe, Mn and Zn in the double mutant, together with the enhanced activity of the Fe uptake machinery, including IRT1 that is known to uptake these metal ions.

4/ Interestingly, LR density increases under Zn excess, primarily due to the severe inhibition of the PR elongation. This prompted us to examine the integrity of the root apical meristem (RAM), which is reduced in the double mutant and shows visible cell death under high Zn.

2/ To address this, we generated frd3 zif1 double mutants and assessed several traits, including developmental parameters and ionomics, across young and mature vegetative, as well as reproductive stages (kinda, stay until the end! 😉)

1/ Citrate and NA have distinct but interconnected functions. a) Citrate supports metal transport in xylem-fed tissues b) NA is involved in intracellular trafficking and phloem-mediated remobilization. Their coordination remains poorly investigated at the physiological level.
