Glucocorticoids (GC), like dexamethasone and prednisolone, are widely used in medicine, but how do they actually work? This is really impressive work (Auger et al.), which figured it all out There are two mechanisms: a slow, broad response, which depends on transcriptional changes by the glucocorticoid receptor (GR) And a second, fast (but sustained) response, where GCs increase flux through macrophage TCA cycles to produce the metabolite, itaconate: Itaconate was produced systemically in response to GCs, and quickly reduced inflammation through a range of mechanisms Here's the most interesting bit: GCs only suppressed inflammation if the TCA cycle and itaconate were in tact, but this was mostly independent of GR's broad transcriptional response - suggesting that the TCA/itaconate pathway is predominant A possible consequence of this: some of the side-effects of glucocorticoids in patients, like increased fat mass and type 2 diabetes, may result from GR's broad transcriptional changes... ... so if we had more precise drugs, which exploit the itaconate response but spare the GR response, this could decouple the beneficial (anti-inflammatory) effects of steroids from the negatives Here it is: nature.com/articles/s4158…

Haussmann Lab ready for #SICB2024 !

Got day 11 and 12 in of the #12WritingDays before Christmas last two days. Today working with past students Shannon McCollum (now PhD student @UMich) and Olivia Canter (now undergrad @DukeU). Writing together on Google docs with the chat feature always fun.

This week did day 8 through 10 of the #12WritingDays before Christmas. Much of it has been with current & past students in the Haussmann Lab (holiday party pic. ❤️ these folks). This type of writing with young scientists is some the most fun and gratifying! #raybucknell #PUI

Today was the 7th day of the #12WritingDays before Christmas! Worked with @Haussben on a paper exploring parent-offspring resource allocation in a long-lived bird species. And, no, not eagles. (Philadelphia Storm-Petrels would be a much cooler team!)

Yesterday was the 3rd day of the #12WritingDays before Christmas where I worked on a storm petrel paper (@BSSkentisland) and zoomed with collaborators Patty Jones and Bob Mauck. Today, the 4th, worked on a dataset from 12 years ago hoping to bring it to the light of day! #PUILife

Yesterday was the 5th day of the #12WritingDays before Christmas & today is 6th! Yesterday joined by @MartineBotany (at culturecoffee) who was putting finishing touches on a paper. I continued to write and give comments on my student @SICB_ posters (methods figure shown)!
