Overall, 2DIR is a fantastic technique that extends the utility of IR spectroscopy applied to hydrogenases and other redox metalloenzymes. Not only can measurements be performed at ambient temperature, it is non-destructive, and high quality spectra are obtained in short time.

3) The terminal COs form a strongly coupled pair where the peaks in linear IR spectra should be treated as antisymmetric & symmetric modes. 4) Computation of anharmonicities is fairly insensitive to the protein surrounding, suggesting these can help predict active site structures

Takehomes: 1) The best structural model is that previously determined with X-ray crystallography. 2) The terminal CN ligands & bridging CO behave as isolated vibrational oscillators. 3) The higher energy CN vibration in the Hinact state is attributed to the CN on the distal Fe.

In this proof of concept study we apply 2DIR to the O2 stable Hinact state of the [FeFe] H2ase from D. desulfuricans. 2DIR allows extraction of intra- and intermode anharmonicities. Combined with computations this provides exquisite insight into active site structure & dynamics.
