📰New & Featured | Can neurons "rust"?🧠 Yes—they can. A fin-loop–like structure in #GPX4 anchors the enzyme to cell membranes, protecting neurons from #ferroptosis. 🧬A Cell study showed that a single point mutation (p.R152H) disrupts this protection, leaving neurons vulnerable to lipid peroxidation—especially in children with early-onset #dementia. Loss of GPX4 triggers #Alzheimer’s-like pathology across multiple models, highlighting ferroptosis as a key driver and potential therapeutic target in #neurodegenerative disease.🧪 Discover the full study here: ow.ly/BUCb50XSLQJ

🚀From Bench to Paper | Hypothalamic Microglia Gate Reproduction via RANK Signaling A new study in Science highlights a pivotal role of hypothalamic microglia in regulating the reproductive axis through RANK signaling, linking neuroimmune control to fertility. 🧠 Loss of RANK—either in microglia or systemically—leads to #HypogonadotropicHypogonadism (HH), with rare RANK variants identified in HH patients. RANK-deficient microglia exhibit impaired activation and structural changes in the median eminence, reducing their interaction with and engulfment of #GnRH nerve terminals. This, in turn, weakens GnRH neuronal responses to #Kisspeptin, a key reproductive signal. 🔬 These findings position microglia as critical regulators of GnRH neuronal function via RANK signaling, with important implications for reproductive development and fertility. MedChemExpress provides #PLX5622 to support this study. PLX5622 in AIN-76A Diet (1200 ppm) enables efficient microglia clearance and is well-suited for studying microglial function in both healthy and diseased states. medchemexpress.com/plx5622-in-ain… #Neuroendocrinology #Microglia #ReproductiveHealth #HPGAxis #Kisspeptin

📚MedChemExpress Digest | Epigenetic Editing Unlike traditional gene therapies, #EpigeneticEditing does not change #DNA permanently. It regulates gene expression reversibly through chemical modifications of DNA and histones, allowing selective activation or silencing of disease-related genes. 🧬Epigenetic editing is a powerful tool for #GeneRegulation, with potential to treat genetic disorders and cancers. Advances in editor design and delivery improve safety and precision, and animal studies show lasting effects in liver, brain, and tumors. ⚠️Challenges remain—understanding epigenetic memory, improving in vivo delivery, and establishing predictive biomarkers. Overall, epigenetic editing represents a shift from “repairing genes” to “reprogramming gene expression”, emerging as a next-generation platform for precision medicine beyond #CRISPR. In this issue, we highlight: 1️⃣ Biological basis of epigenetic editing 2️⃣ Technical platforms & delivery strategies 3️⃣ Applications and clinical prospects 🔗Read more: ow.ly/9JcL50Yw9AT #PrecisionMedicine #HistoneModification #GeneticDisorders #CancerResearch #NextGenTherapies #BiomedicalResearch #TranslationalMedicine

🚀From Bench to Paper | Great to see recently published new work in eBioMedicine investigating how a circulating three-lipid signature (3LS)—previously identified as a poor prognostic #biomarker in metastatic castration-resistant #ProstateCancer (#mCRPC), actively contributes to disease progression. 🌟In this study, MedChemExpress small-molecule compounds were essential tools for mechanistic validation. Researchers used #PF543 (a sphingosine kinase 1 inhibitor) to interrogate the role of #sphingolipid metabolism in driving tumor cell survival. By inhibiting S1P production, PF543 effectively abolished the survival advantage conferred by 3LS-positive plasma, confirming the central role of the ceramide-S1P axis in this phenotype. medchemexpress.com/PF-543.html 🧪Additionally, other MedChemExpress reagents (e.g. #RSL3, #erastin, #ferrostatin1, #BODIPY #fluorescentdye) were used in lipid peroxidation and #ferroptosis-related assays, enabling precise evaluation of oxidative lipid damage and metabolic responses. We are proud to support researchers worldwide with reliable compounds that empower discovery. #LipidMetabolism #CancerResearch #MolecularBiology #Cancer #Oncology #ChemicalBiology

🚀From Bench to Paper | How Sleep Deprivation Impairs Intestinal Stem Cells #SleepDisturbances are linked to numerous chronic disorders, including gastrointestinal diseases—but the mechanism by which the brain communicates sleep disruption to the gut has remained unclear. New research shows that acute sleep deprivation (SD) impairs intestinal stem cell (ISC) function, leading to crypt-villus shortening and Paneth cell loss. The dorsal motor nucleus of the vagus (DMV) was identified as the SD-sensitive brain center transmitting these effects. • SD activates DMV neurons → excessive acetylcholine release from the vagus nerve • Acetylcholine triggers 5-HT release by enterochromaffin cells and suppresses reuptake → gut serotonin spike • Elevated 5-HT induces oxidative stress in ISCs via HTR4 → gut pathologies MedChemExpress provides reagents to support studies on sleep-related gut diseases, including: Serotonin HCl, Prucalopride, SB-203186, GR113808, Hydrocortisone, Mifepristone, Acetylcholine, Atropine, Hexamethonium bromide medchemexpress.com #GutHealth #5HT #SleepResearch #StemCells #Neuroscience #CellStemCell #ResearchTools

🚀From Bench to Paper | Targeting #ATF4 in Elderly KRAS Lung Cancer Age reshapes #KRAS-driven lung adenocarcinoma: slows primary tumor growth but promotes metastasis via epigenetic activation of the integrated stress response (ISR). ATF4 drives epithelial & metabolic plasticity, enabling metastasis through the PERK-eIF2α pathway. Targeting the ISR-ATF4 axis blocks tumor spread and exposes a glutamine metabolism vulnerability. 📈Clinically, high ATF4 in elderly patients links to poor survival—highlighting a promising therapeutic strategy for this understudied group. MedChemExpress supports scientific research. This study made use of multiple MCE products, including #Telaglenastat, #DoxycyclineHyclate, #MG132, and more. #Nature #LungCancer #PERKeIF2α
