Also the SNX9 SH3 binding site in EspF has evolved to prevail in competition. Indeed, residues N-terminal to the consensus +xϕPxxP establish an extended interaction and are essential to strong binding. Therefore, we propose to define the SNX9 SH3-binding epitope as ϕxPxRxAPxxP.

We show that single-repeat EspF can outcompete the autoinhibitory N-WASP C-helix and stimulate actin polymerization. EspF uses C-terminal polar interactions to acquire an EspF(U)-like nM affinity for (N)-WASP GBD.

In this work we characterized the structure, dynamics, and binding thermodynamics of EspF and its SNX9 SH3 and N-WASP GBD complexes.

Our latest studies on the substrate specificities of S. aureus peptidoglycan hydrolases lysostaphin and lytM is now on BiorXiv. biorxiv.org/content/10.110…

We also show that deviating efficiencies towards S. aureus strains with Epr/Lif factors inserting serines into peptidoglycan cross-bridge as well as FemX/A/B deletions shortening the glycine cross-bridge, can be explained by different substrate specificities of these enzymes.

However, hydrolysis of peptidoglycan fragments devoid of D-Ala-Gly cross-linking are generally 5-10-fold slower, suggesting that LytM also targets the cross-linked, mature peptidoglycan.
