📢 Ready to rethink XIST? Our latest work, now in Cell, challenges two fundamental assumptions about XIST: its cis-limited action, and its role in equalizing gene expression between XX and XY genotypes. Dive in 👇🤿(1/6) sciencedirect.com/science/articl…

Excited to share our latest work: L126 fine-tunes enzymatic activity of histone H3, a discovery made possible by viewing the nucleosome as an enzyme. We have a gain-of-function mutant – it’s hard to enhance an activity if it doesn’t already exist! sciencedirect.com/science/articl…

This raises the possibility that autosomal genes in other cell types with a dispersed XIST configuration (pre-implantation embryos, germ cells, immune, lung, and cancer cells) may be regulated by XIST. (5/6)

Applying RAP-seq, we found that XIST spreads across the X and, surprisingly, also to autosomal genes. Autosomal targets show female-specific and XIST-dependent H3K27me3 and downregulation of gene expression, indicating that XIST enforces sex differences on autosomes. (3/6)

In female naïve human pluripotent stem cells (PSCs), XIST is dispersed in the nucleus and X-chromosome dampening (XCD) occurs, raising the question of whether XIST mediates XCD. Knocking out XIST, we found that it is required for XCD in naïve hPSCs. (2/6)

In collaboration with @dinodicarlo, we used hydrogel nanovials to capture and measure the secretion of VEGF-A alongside mRNA transcriptomes. Surprisingly, we found that the VEGFA transcript was not the best predictor of secretion! 2/2

It's time to bring single cell secretion measurement into the multiomics family! Check out our recent BioRxiv submission where we found unexpected relationships between VEGF-A secretion, the VEGFA mRNA, and the cell state: biorxiv.org/cgi/content/sh…

I’m happy to share our new paper with @LabMomoko published today cell.com/stem-cell-repo… which examines how differences in the state of human stem cell pluripotency can lead to favorable or unfavorable outcomes when making brain organoids.

It’s finally out! Many thanks to my colleagues @jacksongcheng and my mentor Andy McMahon at USC for making this happen. #organoid #kidney #screen
USC Stem Cell@USCStemCell

In a @CellStemCell study, scientists from the @USCStemCell lab of Andy McMahon generated kidney-like structures called organoids, and used them to identify potential drugs for adult-onset polycystic kidney disease. @TTdevbio @jacksongcheng @KECKSchool_USC stemcell.keck.usc.edu/mini-kidney-mo…
