@Roseboardman4 Hello, Etching is a pit of an art. Unfortunately it doesn’t look like you got the chelation you wanted. You can trial and error EDTA under varying due actions and pH or try different etching agent: dilute HCl or gluteraldehyde.

Any SEM experts out there please can you help me with my images. (I should see daily increments). This is a glass eel otolith embedded in epoxy resin. I polished until I could see the core with 1/0.05 micron lapping film. I then etched with EDTA and sputter-coated with gold.

@SecorLab Ok thank you for your advice. I will try some different etching techniques. Thank you 🙂.

For reference this is the same otolith under the compound, so increments are definitely present. I just can't seem to get the knack of the SEM. 🫠

In some examples increments are slightly visible but overall I can't use this. I'm getting much better images using a compound microscope.

@clivetrue I don't think so. However some are very crystallised under the SEM. I'm going to try etching for longer to see if that helps! 🤞
