A shot in the dark by our lab to potentially explain species-specific differences and the complexity of the TIGIT signaling pathway, and the colossal clinical failure of TIGIT antibodies. This work was led by two UCDAVIS Undergrads, NOT by PhDs or Postdocs health.ucdavis.edu/news/headlines…

Humans are more cancer-prone than primates despite over 98% genomic similarity. A FasL mutation unique to humans allows plasmin in solid tumors to weaken immune attack @NatureComms * . Could blocking it boost immunotherapy?

*NEW Paper ALERT* Our new work in @NatureComms on FasL-mediated cancer cell death regulation has revealed that evolutionary substitutions (humans vs. primates) in the FasL extracellular domain render human FasL differentially susceptible to plasmin protease cleavage, a key metastatic protease involved in the metastasis of almost all solid tumors including ovarian cancer ascites, etc. Our findings explain the lower cancer incidence in primates (vs. humans) and also the deep mechanistic reasoning behind the potentially less successful outcome of T and CAR-T based immunotherapies against metastatic solid tumors (vs. leukemia, etc). Congratulations to Wamba E, Mondal et al., and all other authors and collaborators @plegembre @carterra_bio (Dan Bedinger) @UCDavisMedCntr @UCD_Cancer @UCD_MMI The manuscript can be found here: nature.com/articles/s4146…

This is the final photograph taken of Hachiko, a Japanese Akita dog renowned for his unwavering loyalty to his owner. Hachiko belonged to Professor Eizaburo Ueno, who resided in Shibuya and taught at Tokyo Imperial University in the early 1920s. Each day, Ueno would walk to Shibuya station accompanied by Hachiko, and they would commute together to his workplace by train. At precisely 3 pm, Ueno would return to the station to head back home, and Hachiko would always be there, patiently waiting to join the professor on their journey. However, one day, tragedy struck when Ueno suffered a stroke and never made it to the station. Undeterred by his master's absence, Hachiko faithfully went to the station every day for nine years until his passing in 1935. To honor his remarkable loyalty, a statue of Hachiko was erected outside Shibuya station in Tokyo. In 2015, on the faithful dog's 80th death anniversary, another statue of Hachiko was unveiled at Tokyo University, symbolically reuniting him with his beloved master, Professor Ueno.
