TrueAllele Investigative Database (TAID) generates and stores DNA genotypes from both simple and complex DNA evidence. TAID also compares genotypes from evidence to other evidence. You don’t need a specific suspect to get a clear picture of crime in your county. As we enter day 27 of investigating Nancy Guthrie’s kidnapping, the investigation seems to have stopped, waiting for the DNA to catch up. DNA mixtures are difficult to interpret because they’re not just one person’s DNA, you’re dealing with multiple possible combinations of the data peaks. Reason 4/4: Math Explosion Every added contributor multiplies the number of possible genotype combinations. At higher contributor counts, manual interpretation becomes harder to justify because there are so many plausible explanations. Case example: Ohio v. Slater Howell III DNA on a white plastic bag was a mixture from seven individuals, so it was sent for TrueAllele genotyping. Testimony described a match between the bag and Howell as 98 thousand times more probable than a coincidental match (and 224 thousand for the victim). cybgen.com/news/cases/Ohi… #DNA #ForensicScience

DNA mixtures are difficult to interpret because they’re not just one person’s DNA, you’re dealing with peaks that could be missing. Reason 3/4: Dropout Real crime scene DNA evidence isn’t clear cut like on TV. Low-level or degraded DNA can cause genetic alleles in the data to “drop out,” so parts of someone’s profile appear to be “missing.” That can make a mixture look incomplete or contradictory. Case example: Glenn Lyons (Reading, PA) A degraded mixture was found on the neckline of a discarded sweatshirt. The lab’s human review reported a match statistic of 42 thousand. DNA mixtures are difficult to interpret because they’re not just one person’s DNA, you’re dealing with different amounts of each person. Reason 2/4: Imbalance Sometimes one person contributes most of the DNA, and another person is just a small amount of DNA. That minor contributor can get lost in the dominant signal, especially when data is limited due to degraded or damaged samples. Case example: State v. Jerod Goodwin A private lab generated DNA data from fingernail scrapings but couldn’t interpret the minor component. DNA mixtures are difficult to interpret because they’re not just one person’s DNA, you’re dealing with untangling multiple profiles stacked together. Reason 1/4: Overlap At each locus, or location in the DNA used for identification, people can share the same alleles, especially relatives. In a mixture, that overlap makes it harder to tell who contributed what and how much. Case example: The Xie “Stain 91” evidence A tiny garage-floor stain contained a complex familial mixture with DNA from at least four of the five related victims.
