A new ray of hope for families facing Hunter syndrome (MPS II). The FDA has granted accelerated approval to Avlayah (tividenofusp alfa-eknm), the first therapy ever designed to cross the blood-brain barrier to treat neurologic manifestations of this rare, progressive disorder in eligible children. For nearly 20 years, there has been no new approved treatment addressing the brain involvement that can cause severe cognitive and developmental challenges. This once-weekly IV infusion was approved based on a meaningful reduction in cerebrospinal fluid heparan sulfate. A confirmatory randomized trial is nearly fully enrolled, and continued approval may depend on confirming clinical benefit. This milestone reflects important progress in rare disease innovation and brain-targeted therapies.
