And, for the love of god... can we stop testing drugs that would be amazing on target solutions for schizophrenia for Alzheimer's and depression??? Or, clinicaltrials.gov/study/NCT00963… is an example of them trialing a partial agonist (already wrong drug) I will say there also probably is a state in disease progression where modulation of NMDA system would not work and maybe my perspective is sub par for pharma bc i think other early synergistic treatments would be best as well.

i understand the lack of emphasis on genetics for something like bipolar, OCD, or depression. but goodness, to think that genetics (along with CLINICAL SYMPTOMS) could be used to intervene EARLY with a devastating pathology is so important yet not used.

schizophrenia is obviously polygenic but here are the main deviations outlined: 1. DAOA overexpression 2. NMDAR abnormalities 3. other genetic neurochemical abnormalities less studied 4. the lifestyle stress that actually leads to the development of schizophrenia. i say this to illustrate that an INNATE independent phenotype exists with slight variability among schizos. but genetics along w clinical symptoms and other tests could be used to identify at risk patients before they develop this disease. how many lives could be impacted!? instead we continue to fiddle around with short sited dopamine blockers post onset which is saddening as there are preventative measures. #schizo #schizolivesmatter

if i were to create a drug for schizophrenia it would be a GluN2A-selective positive allosteric modulator. my add ons would be exogenous allop, 500mg d-serine, and a focus on restoration of gut and metabolic health as to pull down inflammation.

key word is early bc after disease manifestation it is probably too late!

yes being schiz is polygenic but the other ones are instigating factors heeding more neuroinflammation & such. pry sure if u can fix the hypofunc. + pull down neuroinflammation + early therapy many schizophrenics would have just stayed schizotypal.
