First, in the post-surgery setting, the KEYNOTE-564 trial showed that adjuvant pembrolizumab improves not only disease-free survival but also overall survival. Adjuvant pembrolizumab is now a standard of care among eligible patients with clear cell renal cell carcinoma. Second, the TiNivo-2 trial has confirmed, among patients with advanced RCC and who had progression after prior immunotherapy, that rechallenge with tivozanib plus nivolumab is no better than tivozanib alone. This is consistent with prior data from CONTACT-03 and suggests that rechallenge with anti-PD-1 or PD-L1 should be discouraged in RCC. Third, based on CheckMate 67T, subcutaneous nivolumab is now FDA approved as an alternative to IV nivolumab. Fourth, the SUNNIFORECAST trial demonstrated that among patients with divergent histology RCC, nivolumab plus ipilimumab can be an effective treatment option. More results are eagerly anticipated in 2025 and several ongoing trials may continue to change the standard of care in the near future."
