Toxic protein aggregation is a hallmark of Huntington's, ALS, Parkinson's… and enhancing the cell’s natural protein-clearing machinery is a promising therapeutic avenue. @TheVilchezLab at CECAD/University of Cologne studies how protein homeostasis breaks down with age and how stem cells maintain proteome integrity where somatic cells fail. We funded them via @impetusgrants to study toxic protein accumulation and discover ways to reduce those protein aggregates. @TheVilchezLab's Nature Aging paper (supported by @impetusgrants) showed that cold temperature selectively activates PA28γ/PSME3, a proteasome activator required for cold-induced longevity. This reduced disease-related protein aggregation in both C. elegans and human cell models of Huntington's and ALS. His team continues to explore how we might harness these proteostasis mechanisms to prevent age-related neurodegeneration. Glad to support this work!

We also discovered that the deubiquitinase USP4 leads to EPS8 accumulation during aging. Reducing USP4 prevented EPS8 accumulation, extended lifespan, and lessened disease effects. 👉 Targeting EPS8/USP4 could open new therapeutic strategies for neurodegeneration.
