Last day at “From Cells to Organism: When Cell Biology Meets Development” in Paris. It was great to present our new PRIMO OPTICAL PLATFORM, and show live how to photopolymerize hydrogels in 3D using HyPE. They can be used for spheroid and organoid culture, or 3D cell culture.

@cguerin38 @AlveoleLab To challenge the limits of the system, we studied conditions with many beads in the microwells. We then looked at the fraction of weak beads moving, as a function of competition strength (defined by the number of strong beads).

@cguerin38 @AlveoleLab 10. We then demonstrated that when there is turnover, weak structures (from beads or micropatterns) can grow when they are in competition with strong networks. Turnover enables the weakest to exist!

@cguerin38 @AlveoleLab 9. Therefore, without turnover, the actin monomer is limiting and the available monomer pool is taken over by dense structures in a “winner-take-it-all” scenario.

8. To challenge those results in another geometry, the amazing @cguerin38 developed a protocol to generate actin networks with various densities on lipidic micropatterns at the bottom of our microwells (with the Primo @AlveoleLab).
