CONGRESS | #ASH25 | PRESENTATION Samer Srour presents preliminary data of a phase I trial evaluating Orca-Q with (n = 18) or without (n = 26) GvHD prophylaxis (tacrolimus) in patients receiving allo-HSCT with 8/8 HLA-matched donors. Removal of pharmacologic GvHD prophylaxis did not increase GvHD rates and may improve infection rates. At 12 months, cumulative incidence of Grade ≥3 #aGvHD was 8.4% in the prophylaxis cohort vs 5.6% in the without prophylaxis cohort; similarly, incidence of moderate–severe #cGvHD was 11.8% and 0%, respectively. CONGRESS | #ASH25 | PRESENTATION Supriya Gupta presents findings from a phase I/Ib trial of azer-cel, an allogeneic CD19 CAR T-cell therapy + low-dose interleukin-2 (IL-2) in patients with LBCL who relapsed after autologous CAR T-cell therapy (N = 18). Azer-cel + low-dose IL-2 showed a manageable safety profile with Grade 1–2 CRS reported in 72.2% of patients and Grade 1–3 ICANS in 16.7% of all patients. Among the efficacy set (n = 16), the ORR was 81.0% with a CR rate of 43.8%. Robust CAR T-cell expansion was observed among responders (n = 13).
