Pediatric long COVID is characterized by myeloid CCR6 suppression and immune dysregulation Very interesting Spanish study in preprint! #LEONARDI_effect in CHILDREN! ➡️Chemokine receptor CCR6 on the surface of myeloid cells, which include monocytes, macrophages, and dendritic cells, play a key-role in the innate immune system. ➡️Now this study shows CCR6 expression on myeloid cells are SURPRESSED and serves as the most discriminative biomarker for pediatric long COVID, reflecting profound immune dysregulation, including altered innate immunity, lymphocyte overactivation, and impaired SARS-CoV-2 humoral responses, that persists months after acute infection in affected children and adolescents. ➡️This study is highly relevant as one of the first to identify suppressed CCR6 expression on myeloid cells as a strong, discriminative biomarker (79% accuracy) for pediatric long COVID, shedding light on persistent immune dysregulation in children and filling a critical gap in understanding its mechanisms compared to adults. ➡️What this likely means biologically: - The reduced CCR6 on myeloid cells points to a form of innate immune dysregulation persisting months after the initial infection. - It could impair the proper migration or function of these cells to resolve inflammation, contribute to ongoing low-grade immune activation/misbalance (e.g., altered dendritic cell/monocyte subsets observed in the study), or reflect broader exhaustion/altered signalling in the myeloid compartment. - This ties into the study's overall picture of dysregulated innate immunity combined with overactive lymphocytes and weaker antibody responses in affected kids, suggesting myeloid changes (like CCR6 downregulation) as a core driver of why some children develop prolonged symptoms. ‼️Early claims that SARS-CoV-2 poses essentially no harm to children were dangerously oversimplified and wrong, the virus does cause real, sometimes severe or prolonged damage in kids, including acute complications like MIS-C, rare but tragic deaths, and persistent long COVID in a substantial group, with now measurable immune dysregulation that can impair development and quality of life, maybe even life-long! Fasting is making most of you worse and the "mental clarity" you feel is literally a stress response I know this is going to piss people off but I don't care because the science is so clear it's embarrassing… You skip meals. Blood sugar drops. Body doesn't have incoming glucose so it hits the panic button. Cortisol spikes. Adrenaline spikes. Liver dumps glycogen. Free fatty acids flood the bloodstream You feel sharp. Focused. Clean Same neurochemical cocktail as nearly getting hit by a car. Your body is mobilizing emergency reserves because you stopped feeding it. That's all that's happening In a well-nourished person with deep metabolic reserves this is totally fine. Buffer the stress, carry on, no damage But most guys doing 18-hour fasts are already running on fumes. Thyroid sluggish. Cortisol already jacked. Sleep is ass. Digestion compromised. These are the exact people who swear fasting is the best thing they've ever done (because stress hormones feel incredible when you've got nothing else running) and they're the ones being wrecked by it Had a guy come to me doing OMAD for 8 months. Swore by it. Cold hands, terrible sleep, couldn't build muscle, libido completely gone. I told him to eat 3 meals a day for 3 weeks and he looked at me like I was stupid Within 3 weeks his hands were warm and he slept through the night for the first time in over a year. Didn't change anything else Every fast in a depleted system borrows from reserves you don't have. The bill shows up later. Thyroid downregulates further. Gut permeability worsens (cortisol eats the intestinal lining btw, most people don't know this). Hands get colder. Sleep gets worse. Muscle wastes The person who needs fasting the least benefits the most from it The person convinced it's "saving" them is accelerating their own decline Sounds counterintuitive but it's pretty simple once you get it. Stress hormones feel like energy. That's the whole trick. Your body is screaming for fuel and you're interpreting the scream as "wow I feel great without food" Eat the food

@HansAmato For research, simply see animal studies. Animals exposed to one-off fasting (e.g. a 3 day fast) who are then re-fed end up becoming much bigger/stronger than animals that have never fasted. The same dramatic spike in testosterone and other hormones is also seen in humans.

@HansAmato I agree that intermittent/chronic fasting is a terrible idea. But one-off fasting, despite the short-term pain, delivers long-term gains in the form of much higher sensitivity in: - 5AR receptors - Dopamine receptors - Insulin receptors - Lower baseline of inflammation.

@RealAnitaWhite @DonEford @Friesein I actually just saw a report on that from Jefferies yesterday, for the US. Don’t have the global numbers, though. As of Feb. 6th, Novavax has ~227k administrations on the year, which makes up 1.1% of the 20.5M COVID vaccines

In any case, we know the main infection tends to start in the throat/mouth, and only makes its way to the nose a day or two later (human challenge trials). I assume an initial infection is also possible in the lungs. So you need to cover all your bases.

@HansAmato Im starting to think covid attacks the thyroid. I was never vaccinated but got covid in 2022 and my gut and metabolism were destroyed since that point. Im gonna try fixing the thyroid and see where it goes

Most gut issues start with stress-impaired digestion and slow transit time. Low stomach acid, weak enzyme output, and poor bile flow → undigested food → bacterial overgrowth → slower transit → more toxin absorption. And the root of stress intolerance? Low T3 or T3 resistance. Fix thyroid function → become stress resilient → digestion normalizes → inflammation drops → nutrient absorption improves → health peaks. Fix the thyroid, and the gut follows.

@vegetatedgiraf As you know it won't work...simply adding testosterone to your system means you convert it into estrogen (why those on TRT grow boobs and have small balls). Don't think things are bad yet, I'm waiting for cancer to be removed from insurance coverage...for example.

Carrot salad experiment incoming! I'm doing it to see how it's going to affect my estrogen, lipids, liver enzymes, inflammatory markers, thyroid, etc. So basically an in depth experiment as usual. I was thinking of doing 3 carrots per day. What do you guys think? P.S. As you can see by my expression, I'm very excited for this experiment (sarcasm; I'm not a big carrot fan).

In 2020 I published a paper arguing that there likely was an element of CD8 T cell death in Covid. For the following years, I was met with overall antipathy from many professors for claiming covid was harming T cells. Now, a new publication highlights persistent T cell attenuation. sciencedirect.com/science/articl… I was punished the sin of being correct on my matter of expertise before I had done a postdoc or graduated medical school, and for an opinion contrary to many individuals who had incentives to downplay covid or a lack of scientific imagination. My claims were swiftboated, people tried to put words in my mouth, strawman me, and attack me personally. Vincent Raccaniello said the claims covid was harming T cells was "twitter science." Vincent Rajkumar, the editor of Blood, said it was "nonsense." Carlos Del Rio, a professor of Infectious disease said, "I'm exhausted reading such claims from non-immunologists." One immunologist and nephrologist at the Francis Crick institute, Rupert Beale, claimed that cranks were being taken as legitimate sources while I was a medical student. An immunologist at Novartis, Andrew Croxford, mocked me as if I was a self-described 'galileo,' after Zeynep Tufekci referred to me saying, 'not every crank is Galileo.' I'm sorry the truth had to come from someone who lacked pretense, completed fellowships, and several decades of life. It should have been enough to have published in the field, understood the issue, and have been able to articulate the rationale. There were many attacks to my livelihood and my affiliated entities, accusing me of fear-mongering. and writing pseudo-science. One group of Professors even saw it fit to write that the T cells were "Fit and Happy" following breakthrough covid. I have never seen in history someone lampooning an individual with the title of a scientific journal article. Their aim was total demoralization. Having been subjected to this, I believe it is a very unique experience, to be met with derision from 1) Established professors 2) Journalists at the New York Times 3) Journal publications A recent article in the BMJ highlighted how eminent immunologists are starting to believe Covid has harmed population immunity. I was included in the piece until the editor said they needed to avoid the controversy to save face, thereby erasing my advocacy and the absolutely unethical conduct of people who had fixed to attack me. Regardless, this is a significant result. With multiple covid infections, the issue becomes even more relevant. Please keep up to date with vaccinations and avoid covid with use of an N95 if you want to keep your immune system healthy.
