@ATinyGreenCell Fairly large plasmid (lane 2 and 3) about 15.5kb lane 4 9kb plasmid. Classical alkaline lysis and I loaded bit more in lane 3. I am not able to understand what that band is which is marked in RED. Is it ssDNA ? What are those faint bands above the red one?

@DanielaLiebsch @AmodRBhat But we processed are 3 samples at the same time. If the lysis was too long or the neutralization too slow, the extra bands would have been seen in the first sample as well (2nd lane). Not sure that's really the problem here. Btw thanks for the reply!

@AmodRBhat @HemanthGovinde Not sure it is that but sometimes one gets bands of irreversibly denatured plasmid ( lysis too long or neutralization too slow/inhomogenous, esp w midi or maxi. Looks too small for that but u can try if shorter lysis n adding neutr soln fast in 1 go + mix well immediately helps?

@kalapian_ Rather than fighting legal cases, why can't so called "animal lovers" and "influencers" start an animal/dog house. Either crowd fund or invest your money, get "animal lovers" in each city, and start the animal house. It will be helpful to dogs and creates some job opportunities.

1. Wait for written orders of the Supreme Court on stray dogs in NCT. 2. Media is whipping up animal hate frenzy - ignore. 3. I have already reached out to legal professionals who are passionate about animal welfare and rest assured this order will be challenged legally. 4. In the interim, do your best to protect voiceless street animals from animal hating vigilantes who will try to use this opportunity to cause them harm. 5. Animal welfare workers, especially feeders, immediately buy a body camera and ensure you have it ON during your routine feeding to ensure you document any hate/violence towards you. At worst, ensure you have your mobile phone handy to video document any harassment. 6. Don't give up, standing up for voiceless animals is the least we can do in our lives. 7. Don't indulge time wasters & trolls who will try to bait you into useless online arguments. Stay focused.
