@AmodRBhat @HemanthGovinde Not sure it is that but sometimes one gets bands of irreversibly denatured plasmid ( lysis too long or neutralization too slow/inhomogenous, esp w midi or maxi. Looks too small for that but u can try if shorter lysis n adding neutr soln fast in 1 go + mix well immediately helps?

@HemanthGovinde and me, We both extracted this from a 50ml culture and the elute was 1 ml out of which I loaded 5 ul. Help us out.

@ATinyGreenCell Fairly large plasmid (lane 2 and 3) about 15.5kb lane 4 9kb plasmid. Classical alkaline lysis and I loaded bit more in lane 3. I am not able to understand what that band is which is marked in RED. Is it ssDNA ? What are those faint bands above the red one?
