Our newest paper on mitochondrial Topoisomerase 1 shows that Top1mt has only weak endoribonuclease activity and it therefore does not often generate deletions at incorporated ribonucleotides. This helps explain why ribonucleotides are tolerated in mtDNA. academic.oup.com/nar/article-ab…

Our data indicates that the chronic and excessive activation of autophagy contributes to mitochondrial dysfunction. Whether this also drives the age-related mitochondrial decline we will test in a different model system.

We performed a genetic screen in fruit flies and identified several pathways that can rescue a lethal phenotype of mtDNA mutator flies. Looking forward to testing this in a higher species.
