Post 1/2 The paper write up was a historical origin of the invention of Recombinant DNA Methods. This is an old document. I used about 100 restriction enzymes (Table Below) in Gene cloning of 3 constructs in GENBANK including Sanger method Radioactive nucleotide sequencing . Things evolved way past these people's work quickly with the invention of the Polymerase Chain reaction involving my colleague Dr. David Gelfand who Isolated TAQ Polymerase for Kary Mullis. This is not mentioned in the article. Here the restriction enzymes are coded into the ends of the genes or plasmids by building coded DNA primers from synthetic oligos designed with binding Physics in mid and then doing the recombinant work. (Examples shown below). E. coli became a platform for lab protein expression that was very common in Prokaryotes (bacteria). The constructs I created were cloned and protein from ciliates tied to cancer research were expressed, folded, isolated, and characterized. Crystal were grown. However, when your protein expression failed or your proteins could not fold you move to the Lambda viral platforms for gene expression in yeast, etc. The SV40 platforms were BSL Level, because of the cancer risks cited that go back to Judyth Baker's work with Mary Sherman, and others. The paper below that @Kevin_McKernan cited is where the SV40 plasmid construct was used for Eukaryotic protein expression so that characterization and structural analysis with protein crystallography could be performed(cited below Nucleaic Acid Research 1981). The purpose was to be able to express proteins in Eukaryotes (human cells to be specific). With all of this comes risk because when you make mistakes with human cell lines with viruses the outcomes become effectively Bioweapons. Thus the Bio Safety level for such research. Bacterial and Lambda protein expression platforms had real problems making the proper protein structures in eukaryotic cell lines, thus the importance of human cell line transfection like into HELA for protein expression & characterization. On top of that you have DNA code that runs in Prokaryotes and Eukaryotes with the same codon table. The ability to move from bacterial methods human cell lines is a major Scientific advantage in research because the protein translation, folding, and modification is more lake the de novo output of the protein from an in vivo natural chromosome. Berg wanted to answer a fundamental question in molecular biology: Do the mechanisms that regulate gene expression in bacteria also function in eukaryotic (mammalian) cells? He viewed SV40 as an "instrument" analogous to bacteriophages in bacteria—SV40 naturally infects primate/mammalian cells, delivers its DNA into the nucleus, and can either replicate as an episome or integrate into the host chromosome. This made it a promising tool for transduction (transferring genetic material). He planned to: • Insert bacterial genes (e.g., from the E. coli galactose operon, carried on a lambda phage derivative called λdv gal) into the SV40 genome in vitro. • Use the resulting hybrid/recombinant SV40 DNA as a transducing particle to introduce those foreign genes into cultured mammalian cells. • Study whether the bacterial genes would be expressed (transcribed and translated) under eukaryotic regulatory controls, and how gene regulation worked in higher cells. In his 1980 Nobel lecture, Berg explained it this way: He aimed to produce "desired SV40 transducing genomes synthetically" so the recombinant DNA could propagate in animal cells "either as autonomously replicating or integrated DNA molecules." The broader context was his interest in tumor viruses and gene function in mammals, building on his sabbatical work with Renato Dulbecco. We already knew in Science that plasmid constructs in Prokaryotes did this. It was valuable to Science to do this in Eukaryotic cells, especially cancer, for the reasons explained. Like any technology it was a double edged sword, because it could be used in weapon platforms. This ultimately lead to Dr. Titia De Lange's 1994 PNAs paper where the inflammatory function of these viruses lead to cancer cell programming after ~50 cell divisions where the cells either died or became cancer (cited below). These methods have continued to evolve with the automation of computing and robotics leading to safer methods not using radioactivity that use fluorescence. AI has played a major role in strategizing the DNA codes relative to the cloning recombinant methods and the protein expression in the eukaryotic cell lines. Of course mRNA transfection with @RWMaloneMD , and then the modified nucleotide work of recent. Protein structural identification method evolution also occurs.

Post 2/2 So, we get the evolution of a Bioweapon program that we know about from early work. These codes go back millions of years. They are calculated in calculus by the permutation of 2D sequences required to fold into 3D nanomachines. In the case of Coronavirus it is suspected to be ~55 millions of years old originating in birds(cited JVI below). So, like a lot of COTS off the shelf technology the Recombinant DNA Methods got weaponized to levels that are harmful to humanity. We don't have to know all of the reasons why but, we see the trails in US Patents. Like Lyme and the Eric Traub pathogens in ticks this is how we end up here with the SARS Coronavirus Patents and poor applications of technology that hack human DNA Operating systems. The good news is that DNA & RNA are codes that are controlled by a 4bit language and RNA Algorithms that run in Prokaryotic & Eukaryotic cell lines that code for the 20 bit L-Chiral Amino acid based polypeptides with 2 special bits. The Engineers of the codon table made it wobble to preserve function and put in back doors. That is the work of Dr Ethan Taylor and Selenocysteine that can throttle down protein translation kinetics (shown below). The other thing the Engineers gave us were the plants and bacteria made molecules for life. This is where we get Allicin from. It's function we have shown plays it's role in disabling sulfur nanomachine proteins. The DNA Engineers also gave us Abamectin in Prokaryotic bacteria discovered in Japan that can be converted to Ivermectin through catalysis. That molecule inhibits parasitic energy pathways that cancer also leverages. Other research led to synthetic compounds like Benzimidazole was discovered during research on vitamin B12. Fenbendazole was developed in the 1970s as part of a push to improve benzimidazole derivatives. These things all work well together to tackle diseases like coronaviruses, cancer, etc. because they stop runaway hijack of the DNA Operating System and restore normal function. You can never lie in Science. Not collecting data or allowing research is not the Scientific method. Attempting to stop humanity from doing research, even if trial and error is in opposition of true Science. When Science becomes weaponized the bad side of that double edged sword is allowed to evolve. When this type of Sociology occurs humans die. Innocent humans that have no interest in Science at this level are harmed or killed. This is where those of us who do know better work to survive these threats and also create technology to help other humans. The chaos, and the entropy that has been introduced has not helped Science evolve properly. The right thing to do is to have Science benchmarks for all conditions and use our time & materials at the expense of our ability to provide transparent benchmarks in real-time. This may be portals & dashboards. No matter what the way is forward, the days of censoring must end. Humanity must unite in ways of transparent Science benchmarking. Otherwise, the innocent will suffer and this will lead to more death. We are at the fork in the road with technology when it comes to Recombinant DNA Technology and there will be consequences. Making a fiat currency off sick people's lives is a boomerang waiting to hit those that throw it.
