Nat Biotech recently published a provocative study claiming single synapse RNA-seq. Unfortunately, there is no evidence demonstrating droplet-based sequencing of single synaptosomes. They are likely studying random pieces of cellular debris. Our reply: nature.com/articles/s4158…

If I wanna isolate mRNP granules from neurons, what would be the best approach? Transfect cells with GFP-Dcp1a and sort ~500 nm particles? Any other approaches I am not familiar with?
