Goundry (@WillGoRow) and coworkers from AstraZeneca have reported the first and second generation synthesis of TOPO 1 inhibitor AZ14170133 for use as an ADC Payload. They doubled the yield and reduced the PMI by 66%! Check it out here: pubs.acs.org/doi/10.1021/ac…
