I'm very happy to see our 2022 @iGEM project on engineering a probiotic bacterium to tackle gut inflammation finally published in @ACSSynBio! It was the result of an amazing collaborative effort between @UZH_en / @UZH_Science departments.

Anyone know an example of de-novo gene birth from POINT MUTATIONS (not duplications, transposons, fusions), where the gene's activity is also validated experimentally? I only find lists of de-novo genes from bioinformatic screens, or examples from the aformentioned mechanisms...

Fun fact: did you know that ~10% of random DNA sequences have promoter activity? Much more fun to see it directly under a microscope!

@biorxiv_micrbio From the paper they cite doi.org/10.1093/jac/dk… "The results indicate that sertraline may be useful as a model efflux pump inhibitor for in vitro short-term experiments in E. coli, but is unlikely to be clinically useful as a co-drug against Gram-negative bacteria."

@biorxiv_micrbio Sertraline is not approved as an inhibitor of transmembrane transporters, but as an antidepressant. The dose used is 1000x lower than required for efflux pump inhibition! (Note the ng/ml vs. ug/ml ) pubmed.ncbi.nlm.nih.gov/36671340
