Translating Technology: Human Brain Slices as a New Screening Platform Testing neuromodulation technologies such as magnetoelectric nanoparticles (MENPs) in relevant models can be a challenge. Although various in vitro and ex vivo platforms such as cell cultures, rodent brain slices, and organoids offer alternatives for screening neural implants before in vivo studies, each still falls short of reproducing the full structural and functional complexity of human brain tissue. New research by Eggert et al., published in the Journal of Neural Engineering, is helping advance this field by establishing organotypic human brain slice cultures as a screening platform for neuromodulation techniques. Using CorePlate™, the team demonstrated spiking and field‑potential activity is present in fresh human brain slices treated with MENPs. Offering a high‑resolution, human‑relevant method which can help develop ideas from engineering concepts to clinical applications, providing an ethically preferable and early‑stage alternative to animal models for evaluating emerging neurotechnologies. Mapping the Hidden Mosaic While the vertebrate retina is typically organized into distinct layers, displaced retinal ganglion cells (dRGCs) are different in that their cell bodies lay within the inner nuclear layer rather than the ganglion cell layer. The question as to the true role of these dRGCs, and if they are integrated members of the visual circuit has been a question in retinal neurobiology for quite some time. New research by Sabrina Duda et al., alongside Christian Puller & Martin Greschner published in Scientific Reports, is improving our knowledge of dRGCs by mapping the spatial distribution, function, and receptive fields of these unique cells, and finding out how they fit into the sustained ON alpha (sONɑ) RGC mosaic. Utilizing CorePlate™ enabled HD-MEAs with 4,096 simultaneously recording electrodes, the group was able to capture the electrical fingerprints of dRGCs alongside their regularly placed counterparts. By correlating their light-evoked responses, they demonstrated that displaced sONɑ RGC receptive fields fit into the gaps of the regular cell mosaic and serve the same functional role as others within the mosaic. This research reveals that cell body position can be surprisingly flexible and shows that even "displaced" neurons are essential components of the brain's complex sensory circuits. Excited to see this ALS research move from preprint to publication! The newly published work from Vegard Fiskum et al. in Network Neuroscience offers powerful new insight into how motor neurons are affected in ALS. By combining human‑derived cell models with advanced functional analysis from CorePlate™ enabled HD-MEAs, they found that ALS neurons were hyperactive with smaller spike amplitudes. Further to this, they found that the ALS neuronal network was affected, showing an increased reliability on a small group of highly interconnected neurons to maintain function (increased rich club effect). Studies like this bring us closer to understanding, and ultimately treating this complex disease.
