What an incredible experience at the 4th Next-Gen Organ-on-Chips & Organoids Conference! We were proud to sponsor such an event and connect with so many researchers that put their effort into pushing the boundaries of organoid-based research. Particularly intriguing were the advancements in personalized drug screening using patient-derived iPSCs, which hold great potential for tailored treatments and disease modeling. Are we ready for in vitro cognition? Biological AI delves into the possibility of using biological models like neuronal cell cultures and organoids to emulate human cognition, potentially training machine learning models. But what if computers themselves were biological? Biological AI pushes the boundaries of traditional computing by exploring the possibility of using human-brain-based circuits as alternatives to silicon chips. @NeuChiP is a project that promotes human stem cell-derived biological artificial intelligence. This cross-disciplinary consortium explores biologically trained machine learning models, as well as engineered biocomputers. Microglia regulate neuronal development and synaptic refinement through Trem2 Recent research highlights the #immunesystem's role in #CNS development and ageing. Trem2, expressed exclusively in the #brain by #microglia, modulates energy metabolism and enhances phagocytosis. Variants on Trem2 are linked to neurodegenerative diseases like Alzheimer's. However, its role during neurodevelopment remains unclear. In this study, @EricaTagliatti et al. demonstrate that Trem2 deficiency disturbs neuronal transcriptomic and energetic profile during development, causing altered connectivity and behavioral defects in adult mice brains. Tagliatti and colleagues exploit 3Brain’s CorePlate™, a microchip-based technology to record and analyze high-content functional activity. Analysis on hippocampal brain slices in Trem2-/-mice revealed synaptic and functional alterations in the CA1 region but not in CA3. These findings, along with others, strongly suggest that the synaptic deficit in adult Trem2-/- mice specifically occurs in the CA1 region. This study opens the door to explore whether reduced neuronal activity in aging and neurodegeneration correlates with Trem2 deficiency Congratulations to @EricaTagliatti and colleagues. Creating 3D models with #brainorganoids is invaluable, yet they present challenges: - Long protocols for organoid creation, often with single-use testing. - Surface activity recordings offer limited insights. The Accura 3D HD-MEA overcomes these challenges. Its thin microelectrodes penetrate organoids, capturing deep dynamics and interactions to provide accurate information about density connections in a 3D environment. Additionally, these microelectrodes are non-destructive to organoid’s cytoarchitecture, allowing for prolonged use in long-term studies.
