preprint Alert📢 Time to talk about the🐘 in the room of genetically encoded Calcium indicators (GECIs). These indicators are widely used to report activity in the brain and an invaluable tool, but the expression of these GECIs can change activity. 🧵1/7 biorxiv.org/cgi/content/sh…

Yesterday, I was so happy to find nice neurons in the visual cortex for Ca2+ imaging, and I could selected them using chessboards in 3D...but during the measurement, the mouse started to run... I needed to find a good solution to avoid movements! (coming soon) #neuroscience #AOD

⚠️Weather update⚠️: after our observation of the hippocampal storms🌩️, our neurometeorologists started analyzing the phenomena. First, we measured the speed of the waves, which were around 24.4±3µm/s. Do you have an idea on what could spread so slowly? #hippocampus #neuroscience

@4Specialists I saw this wave too, using UCLA miniscope and the same virus in the hippocampus, the mice were freely moving, on the linear track. At first, I thought this wave related to the behavior, but it was not. So I don’t know how to explain it.😭

Some details: Video was taken by 2p acousto-optical microscope. Scanning freq ~40 Hz, on 500x500 um. AAV GCamp6f was injetced in the HC of mouse. During experiment the headfixed mouse was running in a gramophone.

@Noni_sdh Thanks for the comment! It looks overexpressed indeed. However we have never seen similar slow waves before! It was recorded by 2p AO microscope from @Femtonics.

@4Specialists Overexpression of GCaMP. Try to dilute the virus and lower the LED power. If you are working with these mice for a long period, you need to find the minimal dilution that produces a decent fluorescent signal after ~4 weeks.
