$PRME PLAYED CHESS AND WON When Prime Medicine announced in March 2025 that it would develop an AATD candidate, the market read it as a mistake: why enter such a crowded field, and against a company it shares scientific origins with? The correct reading was the opposite. This was neither defensive nor opportunistic, it was a calculated strategic move, and today’s ruling confirms the game has been won. The real prize isn’t an indication. It’s the opportunity to demonstrate superiority against the entire field of genetic medicine and to do it on the most closely watched stage there is. AATD is the perfect proving ground for two reasons. First, because the disease demands solving two problems at once: clearing the toxic Z protein that accumulates in the liver and restoring the functional protein that protects the lung. Knockdown strategies (siRNA) address only the liver; augmentation therapy, only the lung. Correcting the mutation at its source solves both in a single step. Second, because within gene editing itself, PM647 tests the precision of prime editing against base editing in a comparison with no noise: same gene (SERPINA1), same mutation (E342K / Pi*Z), same organ, same vehicle (universal LNP). When you isolate a single variable, you build the cleanest experiment possible. And $PRME doesn’t arrive empty-handed: in fully humanized mouse models, PM647 restored the corrected protein (M-AAT) to the healthy human range at clinically relevant doses. A second potential victory follows from that experiment. If prime editing wins on an identical target, it stops being a promise and becomes an extrapolable read-through: wherever both modalities overlap, $PRME starts with the edge over $BEAM. And it’s worth remembering that both technologies were born in the same lab, prime editing is the evolution of base editing, and winning on its own turf is the most direct way to prove it. The legal-strategic one. Entering AATD forced a tribunal to draw the boundaries of the 2019 collaboration agreement. The ruling doesn’t just secure PM647 (within Prime’s “Field,” with no damages owed to Beam), it confirms the operating perimeter of the entire platform. What was an existential risk, the possibility of having to hand over or abandon the program, becomes clarified freedom to operate. This isn’t a bet on a program, it’s a bet on the platform. Wilson (PM577a, already cleared for the clinic) and AATD (PM647, IND/CTA in Q3 2026) both advance on the same universal liver LNP. Validate one clinically and you validate the vehicle for the other and for every future liver target. The market keeps valuing program by program when it should be valuing the whole platform. The “saturation” that looked like a threat was, in fact, the strongest card: winning where everyone is watching counts for far more than winning where no one compares. Entering AATD was never the risk. It was the move. Today the last piece falls into place. Initial clinical data in 2027.
