PROGRAMMABLE BACTERIA IS KEY IN THE WIRELESS CONTROL OF MOLECULES USING FREQUENCIES Biological Computing 3.1.3.5 Plasmid DNA Plasmids are one of the indispensable common carriers in genetic engineering, perfectly meeting a series of conditions required as a genetic engineering carrier [7]. Plasmids are essentially a sub-cellular level genetic element, neither wrapped in a protein shell nor having an independent life cycle outside the cell. It depends on the host cell for replication & proliferation, stably inherited to the daughter cells w/ the division of the host cell, but once it leaves the host cell environment, the plasmid cannot survive independently. Plasmids have a certain compensatory effect on the function of the host cell, & they achieve this by regulating various biological processes. Plasmids have the ability to replicate & transcribe autonomously, which is one of the key characteristics of them as gene carriers. They can ensure a constant copy number in daughter cells, thereby stably transmitting & expressing the genetic information they carry. In addition, the existence form of plasmids in cells is flexible & diverse, they can either independently exist in the cytoplasm or integrate into the bacterial chromosomal DNA, this flexibility provides great convenience for genetic engineering operations [7]. As stable genetic elements outside the chromosome, plasmids typically range in size from 1 to 200 kilobase pairs (kb), presenting a double-stranded closed circular DNA structure. In terms of structural complexity, plasmids are relatively simple, even more so than viruses (lab made). In E. coli, a model organism, scientists have identified & classified various types of plasmids, the most well-known of which include F plasmids, R plasmids, & Col plasmids. F plasmids, also known as F factors or sex factors, have a unique ability to carry genes from the host chromosome & transfer them to recipient cells that originally did not contain the plasmid, achieving horizontal gene transfer. R plasmids, known as resistance factors because they encode one or more antibiotic resistance genes, not only endow bacteria w/ resistance to specific antibiotics but can also transfer this resistance to recipient cells lacking the corresponding plasmid under suitable conditions, enabling the latter to also acquire antibiotic resistance. As for Col plasmids, they are a type of factor that produces colicins, the genes they encode control the synthesis of colicins. Colicins are a type of protein w/ strong antibacterial activity, capable of killing bacterial strains that are closely related to the E. coli producing the Col plasmid but do not carry the plasmid. In the coding design of plasmid DNA, all plasmid DNA molecules suitable for use as gene cloning vectors share 3 core components: Replicon structure, Selective Marker, & Cloning Site. Specifically, the Replicon structure is the basis for plasmid self-replication, which includes a key replication initiation site (Origin of Replication, abbreviated as ori) responsible for initiating the DNA replication process; it ALSO CONTAINS REGULATORY GENES TO CONTROL THE FREQUENCY & EFFICIENCY OF REPLICATION, & Replicon coding genes, which are crucial to the plasmid’s replication mechanism. The Multiple Cloning Site (MCS) consists of a series of single restriction enzyme cutting sites, providing precise insertion points for exogenous DNA fragments, allowing researchers to insert target genes or DNA sequences into specific locations on the plasmid as needed. link.springer.com/content/pdf/10… Building machines w/ DNA molecules We discuss the potential of DNA for creating machines that are both encoded by & built from DNA molecules. Everyone celebrates Bitcoin volatility declining. 🤦‍♂️ -87% → -84% → -77% → -45% But that's not what matters. Look at the market cap wipeout: -$21B → -$350B → -$1.13T → -$1.33T Today's Bitcoin is a completely different asset than it was in previous cycles. From a capital allocator's perspective, surviving a $1.3T wipeout is not the same game as surviving a $21B one. Only when #BTC successfully comes back from this — can we definitively say it survived. And that's the real test. Not the % drop. The absolute destruction of wealth it can absorb and come back from. 👇🤝
