We observe an exponential increase in mtDNA deletion frequency with aging, as well as increase in the mean deletion size with age. While we observe similar frequency of deletions between tissues, sequencing suggests a tissue specific distribution of deletion breakpoints.

@AmyLeMessurier @Momademia I think in retrospect I wouldve used the early months with frequent pumping to prioritize things with more flexibility (analysis, etc) and remember that it’s a short time and you’ll space pumping more as time goes. Try to be kind to yourself, it’s a super human endeavor!

how do people manage doing experiments with needing to pump every few hours? (Without spending super long days in the lab?) Breastfeeding is starting to feel incompatible with lab work 😥@Momademia

@AmyLeMessurier @Momademia Such a challenge early one I’m sorry you’re dealing with it. I would try to have a pump space immediately adjacent to lab, assemble pump parts in the am and store them in fridge between sessions, and then do short sessions focused on comfort when necessary to get a protocol done.

These data provide invaluable information for future work to understand the mechanisms underlying formation and amplification of mitochondrial genome deletions and for using these variants as a metric of aging. Thank you to @timp0 for collaboration!

Applying our method to human brain substantia nigra tissue, we identify a similar frequency of mtDNA deletion events to that observed in muscle for both younger and older samples, however we observe a distinct spectrum of deletion locations.
