Botox in the face may reduce orgasm intensity in women, and it can affect depression scores depending on where you get injected. (PMID 30283017) 36 women: 24 got actual Botox and 12 got other cosmetic treatments as controls. A significant drop in the orgasm domain for the Botox group was found on the Female Sexual Function Index. No change in the controls. "Reducing the ability to make the facial expressions associated with sexual pleasure leads to a reduction in the reported feeling of pleasure associated with it. The results suggest that the facial expressions do not occur simply to communicate pleasure to a partner but they are an integral part of the feeling of pleasure and are important in the process of achieving orgasm." The results for depression went in two different directions: Injecting the crow's feet (laugh lines) worsened depression scores. Injecting the glabellar frown lines improved them. Small sample size, and these findings haven't been replicated or refuted yet. I still think this is enough to make you think twice about Botox in the upper face, especially frown lines, crow's feet, and laugh lines. More research is needed, but this is pretty concerning.

The mouse amino acid literature is a minefield. Restrict glycine and alanine for better weight loss. Restrict bcaas and methionine to reduce mTOR centric aging. Reduce arginine in some cancer types. Less Isoleucine for better body comp/metabolic function Other examples, too. What is one to do?

In obese mice under 40% calorie restriction, higher glycine intake vs alanine control led to more favorable weight loss: - 14% more fat loss - 26% lower fat depots - 27% less lean mass loss (during early phase) - 4% greater muscle preservation Both groups were calorie restricted to the same level, but the glycine mice lost slightly more fat and slightly less lean/muscle mass. Imagine a drug for reducing depression or anxiety that is as powerful as Tirzepatide is for reducing hunger. I think tens of millions of people will be on drugs like this in the next 5 years and I think it will be incredible. I think we’ll look back on the rest of history when such medication was yet to be available and find it very brutish.

These are really really nice data that've gone largely under the radar In people already taking a statin, Enlicitide (an oral PCSK9 inhibitor) was tested head-to-head against other cholesterol medicines — Bempedoic Acid, Ezetimibe, or the combination Enlicitide lowered LDL more than even the combination (and was the only agent to lower lipoprotein(a) too, by ~25%)
