@andrewcaravello I can’t get over how everything you write is so perfect to read! You have brought a civility to the NWBO discourse that had disappeared. Even the grumpiest people have softened their tone or disappeared. Thank you!

$NWBO We all came into this at different times. Some of you have been here longer than I have. Others are just getting started. I’m probably somewhere in the middle. But I think right now, more than anything, we need to zoom out. This isn’t the moment to worry. This is the moment to pause, take a breath, and let the process play out. Everything is already shifting. Not in theory, but in real time. It’s happening because something this big doesn’t move the same way as everything else. The system is already bending around it. I don’t spend time anymore wondering if this will get approved. I believe it’s already decided. I just don’t know the day. But I would bet my reputation that it’s coming. In the meantime, I’ve actually enjoyed digging into the science behind it all. I’ve been using ChatGPT to help explore the different parts of the platform, and it’s made things a lot easier to break down. I appreciate you all sticking with me, especially when I get things wrong. I’m just a person like anyone else. I make mistakes. But I always try to get it right. What still hits me the hardest is knowing that every day this is delayed, another patient goes without something that could help them. That’s what keeps me going. It’s not about a ticker. It’s about a treatment that could change someone’s life. And yes, we live in the real world. A messy one. Governments argue. Companies compete. Even people trying to change the world can’t always get along. But we’re still here. Still watching. Still waiting. And I think we’re closer than ever.

$NWBO 🎯 Can DCVax-L Be Used to Identify Precision Medicine Targets? Yes—and here’s how. DCVax-L isn’t just a therapeutic—it’s a personalized immunologic mirror of a patient’s tumor. As such, it inherently reveals what the immune system can recognize, has failed to recognize, or might be re-trained to recognize. Let’s break down the target discovery potential of the DCVax-L system into actionable phases: ⸻ 🔬 Step 1: Antigen Processing by Autologous Dendritic Cells DCVax-L is made by pulsing autologous dendritic cells with a patient’s own tumor lysate. 🧠 That lysate contains: •Patient-specific tumor neoantigens •Clonal and subclonal mutations •Tumor-associated self-antigens 🧬 These antigens are then processed via: •MHC Class I (CD8+ T cell priming) •MHC Class II (CD4+ T cell support) ✅ The dendritic cell response reveals the immunologically relevant antigens—those that actually provoke T-cell activation in humans. ⸻ 🔁 Step 2: Immunopeptidomics + Single-Cell Immune Profiling Using post-vaccination T cells (or from in vitro DC-T co-cultures), one can: •Perform TCR sequencing to identify dominant clones •Use mass spectrometry to identify peptides presented by MHC •Cross-reference tumor mutanome with the actual immunopeptidome •Apply single-cell cytokine or cytotoxicity assays to prioritize targets 🎯 Result: You obtain a shortlist of actionable targets that: •Are processed and presented in real patients •Are recognized by T cells •Can be stratified across populations (e.g., shared across glioma patients) ⸻ 🤝 Could This Be Franchised? Yes—and NWBO is in a uniquely strong position to commercialize this model as a franchisable discovery platform. Here’s how that could work with a partner like Amgen: ⸻ Example Partnership Structure: NWBO x Amgen NWBO Provides: •Access to Flaskworks-powered DCVax-L manufacturing •Rights to use patient-derived tumor lysate + DCs for discovery •Flaskworks systems franchised to academic centers or CROs Amgen Does: •Runs multiplex immunopeptidomics and AI analysis •Develops biologics (e.g., BiTEs, ADCs, or off-the-shelf vaccines) •Conducts multi-center trials based on newly discovered targets Outcome: •Validated target peptides for ADCs or T-cell engagers •NWBO collects milestone + royalty •NWBO leverages global deployment w/o internal clinical ops overhead ⸻ 💸 What Does NWBO Capture Financially? Source: •🔁 Upfront access/usage fees •💼 Milestones upon entering clinical trials or achieving ORR endpoints •📈 Royalties on any FDA-approved drugs targeting epitopes discovered via DCVax-L •🔬 Platform equity via optional joint ventures for co-owned targets ⸻ 🧠 Strategic Advantages NWBO Has Over Pure-Play Target Discovery Firms 1. Real-World Immunologic Response Generation DCVax-L reveals what the immune system actually recognizes—not just computational predictions. 2. Tumor-Agnostic Personalization DCVax-L doesn’t rely on a fixed target—it uses the entire tumor lysate. Every patient = unique dataset. 3. Flaskworks Automation Sensor-integrated, cartridge-based DC production enables standardization, reproducibility, and scalability. 4. Deep Immune Learning Reservoir NWBO has decades of patient-derived data from GBM and solid tumors—making them a potential Rosetta Stone of T-cell recognition in cancer. ⸻ 🚦Regulatory Angle: Why FDA Would Support This Model The FDA is now: •Supporting platform designations •Encouraging non-pharma target discovery and biological response profiling •Promoting public-private partnerships, especially where novel CMC systems (like Flaskworks) lower cost and boost reproducibility This positions NWBO not just as a therapy provider, but as a hub of immunologic data and target discovery. @ClankoMedia @X_Is_Lies @HockeyThinkTank @MYHockeyRanking I think my tone has been pretty positive! I don’t work for them but I do think what they created has done more good for non-professional hockey than any USA Hockey policy or any youth hockey ‘experts’ who promote themselves and their products. Not sure why X_Is_Lies is so mad!

@X_Is_Lies @Brian_Moore21 @HockeyThinkTank @MYHockeyRanking “Dude” There’s no script. They have never once come out and said that their rankings are the Bible. It’s not their fault that various hockey organizations/entities have taken their rankings and saw fit to use them for other purposes like seeding and at large bids.

Dear Youth Hockey, Today is the day. It's an important day. A day in youth hockey that everyone's been waiting for. Today, @MYHockeyRanking has released it's first rankings! I've already gotten a message from a hockey director about an angry parent about their team's ranking. And I'm sure it won't be the last. I've already seen the posts from programs highlighting their teams rankings on social media. Who cares. It's all marketing BS. Coaches should be judged based on the type of practice that they run, not the ranking of their team. Development should be the watermark, not recruiting. I was a college coach and I never looked at the ranking of a team when I was recruiting players. It didn't matter. And it never will. To be honest, if I talked to a parent or a coach and they gloated over it...it would be a red flag. So let's relax. Let's let the kids have fun playing the game they love. Let's challenge them in practice to be better. Let's develop the life skills in these kids that matter. And let's quit it with what I believe is the thing that is causing so much angst and uncertainty in our kids today- comparison. The kids deserve better from us adults. Let's be better.

@X_Is_Lies @ClankoMedia @HockeyThinkTank @MYHockeyRanking Nobody’s even mentioned the players, who like to know the teams they’re playing, like to follow their buddies who move to other teams, and simply enjoy following teams in other leagues. I’ve yet to meet a kid who doesn’t understand the intuitive concept of MHR.

@X_Is_Lies @ClankoMedia @HockeyThinkTank @MYHockeyRanking You can’t fake your way to the top of the rankings. The most competitive hockey is played by the teams at the top. What’s wrong with everybody seeing that? Coaches can’t game the system, and if they’re not developing kids the numbers will show it.

@X_Is_Lies @ClankoMedia @HockeyThinkTank @MYHockeyRanking Wrong. Parents want to play appropriate games. The disconnect is when teams that are an 86 beat up on teams that are a 77 or enter a tournament against teams that are a 93. A coach of a team that is an 86 will never get a complaint by playing teams in their range—84-88.

@Brian_Moore21 @ClankoMedia @HockeyThinkTank @MYHockeyRanking Missing the point - associations and parents equate these rankings with worth. Not one parent looks at the rankings solely through the lens of “how should we schedule games?” I mean, have you met hockey parents? Being in a “winning” association very often trumps development.

@HockeyThinkTank @MYHockeyRanking MHR is a scheduling tool and nothing more. They are proud of that, as they should be, because it helps ensure competitive games. But also, nothing wrong with marketing your team’s ranking, any more than it is marketing your facilities, history of success, coaches etc.

@HockeyThinkTank @MYHockeyRanking I can’t believe you had on a guest whose product you said you didn’t like and neither one of you did any research. I was embarrassed to be listening to it. Five minutes of research would have allowed you to ask thoughtful questions. Neil handled the disrespect very well.
